摘要
建立和验证了核桃渣中单宁酸物质的高效液相色谱检测方法。通过对色谱条件的摸索和优化,采用Inertsil ODS-SP C_(18)(250 mm×4.6 mm)色谱柱,以2%冰乙酸和乙腈作为流动相梯度洗脱分离单宁酸物质,流速为1 mL/min,检测波长为280 nm。结果显示,各物质呈现良好的线性关系,加标回收率在80.66%~108.17%,重复性、稳定性和精密度均小于10%。该方法的灵敏度高、操作方便、准确性良好。
An efficient high-performance liquid chromatographic gradient method for the determination of tannins in walnut residues was developed and validated. The chromatographic conditions of tannins were investigated and established. The methanol solution was chromatographed on an Inertsil ODS-SP C(18)(250 ×4.6 mm) column, with a gradient of mobile phase: 2% acetic acid glacial-acetonitrile, the flow rate of 0.5 mL/min and tannins was detected at 280 nm.The result showed that the method had a good linearity, the recovery rate was between 80.66% and 108.17% ,and the RSD of repeatability, stability and precision were all lower than 10% . The method is high sensitivity,conveniency and accuracy.
