摘要 采用浓缩胶浓度为5%,分离胶浓度为12%的SDS-PAGE凝胶系统,运用Image J软件分析不同蛋白质浓度、pH、NaCl浓度对肌原纤维蛋白的分离效果。结果表明,在蛋白质浓度为1.2 mg/m L、pH为7.5、NaCl浓度为0.5 mol/L时,肌原纤维蛋白中各蛋白质及其亚基实现较好分离,均匀分布于整个电泳条带,且各蛋白条带光密度值能清晰分辨。Image J软件能有效地应用于SDS-PAGE图像分析。 To investigate the separating effects of different protein concentration, pH and NaCl concentration on myofibrillar protein extracted from grass carp (Ctenopharyngodon idellus),based on SDS-PAGE(5%,12%) and Image J software,the sepa- rating effects of different protein coneentration,pH and NaCl concentration on myofibrillar protein were analyzed. The results showed that when protein concentration was 1.2 mg/mL,pH was 7.5 and NaC1 concentration was 0.5 mol/L,the proteins and their subunits of myofibrillar protein could be well separated and evenly distributed in the whole electrophoresis band,further- more the optical density of each band could be clearly distinguished. Therefore,Image J software could be usefully applied on SDS-PAGE image analysis.
