摘要
在育种后代材料中发现1株水稻(Oryza sativa L.)颖壳扭曲突变体P303,遗传分析表明,突变体表型受1对隐形基因控制。根据TWH1基因的初步定位结果,利用F2定位群体对该基因进行精细定位。结果表明,TWH1基因被定位在RM14148和RM14167之间的0.6 c M区间。通过在目标区间设计SSR引物,TWH1基因最终被定位在SSR标记RML25和RML35之间,物理距离为11.9 kb。基因注释表明在该区间只有1个候选基因LOC_Os02g56610,该基因编码一个包含DUF640结构域、全长248氨基酸残基的蛋白质。序列分析表明,突变体在该基因第二个内含子区域发生了17个碱基的缺失,从而导致开放阅读框的改变。
A mutant P303 with twisted hulls was found in a breeding population of rice (Oryza sativa L.). Genetic analysis indicated that the mutant phenotype is controlled by a single recessive gene. Using a map-based cloning strategy,the TWH1 gene was narrowed down to a 11.9 kb region between the SSR markers RML25 and RML35 on the long arm of chromosome 2. Sequence verification revealed that the twhl mutant harbored 17 bp deletion in exon 2 of LOCOs02g56610 which resulted in a frame-shift mutation. LOCOs02g56610 encodes 248 amino acids and contained a DUF640 domain-like gene.
