摘要
目的探索健脾益气摄血方及其拆方对辛伐他汀诱导斑马鱼脑出血模型的防治效果。方法将660尾受精后1 d(1 dpf)黑色素等位基因突变型Albino品系斑马鱼随机分为正常对照组、模型对照组和实验药物A、B、C、D组及其分别下设100、250、500、750、1 000 mg/L不同浓度组,每组斑马鱼30尾。实验过程在严格盲态下进行,同时加入不同浓度的实验药物A、B、C、D和0.5mol/L辛伐他汀处理斑马鱼24 h,每组选取20尾斑马鱼镜下观察出血情况,拍照、观察斑马鱼脑出血尾数和出血发生率(S)计算出血减少率。结果 (1)健脾方100、250、500、750 mg/L浓度组出血发生率分别为75%、70%、60%和70%。(2)健脾益气方100、250、500 mg/L浓度组出血发生率分别为60%、40%和85%。(3)健脾益气生血方100、250 mg/L浓度组出血发生率分别为50%和45%;出血减少率分别为29%和36%。(4)健脾益气摄血方100、250、500 mg/L浓度组出血发生率分别为35%、25%、45%;出血减少率分别为50%、64%和36%。结论健脾方和健脾益气方对辛伐他汀诱导斑马鱼脑出血无防治效果;健脾益气生血方与健脾益气摄血方显示有良好防治效果,但高浓度具有毒性。
Objectives To explore the preventive and therapeutic effects of Jianpi Yiqi Shexue Keli( Spleen-fortifying Qi-tonifying Hemostatic Granule,JPYQSXKL) and its disassembled prescriptions( Jianpi Granules) in the treatment of zebrafish cerebral hemorrhage induced by simvastatin. Methods 660 melanin alleles mutant Albino zebrafishes were randomly divided into normal control group,model group,Jianpi groups( A,B,C,D) and subgroups of their different concentration( 100 μg/m L,250 μg/m L,500 μg/m L,750 μg/m L,and 1 000 μg/m L)( n = 30 in each group). Blinding measures were strictly followed throughout the process. The model of intracranial hemorrhage was established by adding 0. 5 μM simvastatin while administering granules of different concentration at the same time. 24 hours later,20 fishes out of each group were observed under the microscope to count the number of hemorrhagic fishes( S) and thus calculate hemorrhage percentage and its decrease rate. Results(1)The incidence rate of hemorrhage in Jianpi groups( 100 μg/m L,250 μg/m L,500 μg/m L and 750 μg/m L) was 75%,70%,60% and 70%;(2)The incidence rate of hemorrhage in Jianpi Yiqi groups( 100 μg/m L,250 μg/m L and500 μg/m L) were 60%,40% and 85%;(3)The incidence rate of hemorrhage in Jianpi Yiqi Shengxue( Spleen-fortifying Qi-tonifying Blood-nourishing) groups( 100 μg/m L and 250 μg/m L) were 50% and45%;(4)The incidence rate of hemorrhage in Jianpi Yiqi Shexue groups( 100 μg/m L,250 μg/m L and500 μg/m L) were 35%,25% and 45%. Conclusions No protective effects of Jianpi granule and Jianpi Yiqi granule on intracranial hemorrhage induced by simvastatin in zebrafish were observed. Jianpi Yiqi Shengxue granule and Jianpi Yiqi Shexue granule appeared to have a good effect yet high dose was toxic.
