摘要
目的探讨益坤抑痛平颗粒缓解子宫腺肌病患者痛经的可能作用机制。方法 30只ICR小鼠随机分为空白组、模型组和中药组,每组10只。模型组和中药组以口服枸橼酸他莫昔芬片法建立小鼠子宫腺肌病模型。造模成功后空白组和模型组以生理盐水10μl/(g·d)灌胃,中药组以益坤抑痛平颗粒混悬液15μl/(g·d)灌胃,均给药8周。肉眼观察各组小鼠子宫形态并称量子宫重量,检测各组小鼠子宫内膜中神经生长因子(NGF)及其受体P75的表达。结果空白组小鼠子宫表面光滑,子宫表面未见异常结节;模型组和中药组小鼠子宫表面可见腺肌病结节。中药组和模型组小鼠子宫重量均重于空白组(P<0.05);而中药组与模型组小鼠子宫重量差异无统计学意义(P>0.05)。各组小鼠NGF蛋白表达差异均无统计学意义(P>0.05)。模型组小鼠子宫内膜中P75表达高于空白组,而中药组小鼠子宫内膜中P75表达较模型组降低(P<0.05)。结论益坤抑痛平颗粒可能通过降低子宫内膜中NGF受体P75的表达来达到缓解子宫腺肌病痛经的目的。
Objective To explore the possible mechanism of Yikun Yitongping Keli( 益坤抑痛平颗粒) in relieving adenomyosis( AM) patients' dysmenorrheal. Methods Thirty Institute of Cancer Research( ICR) mice were randomized into blank group,model group and herb group,10 mice in each group. Mice in the model group and the herb group were made AM models by oral tamoxifen citrate tablet method. After successful modeling,the blank group and the model group were given normal saline 10 μl/( g·d) by gavage. The herb group was given Yikun Yitongping Keli suspension 15 μl/( g·d) by gavage. All groups were treated for 8 weeks. Uterine morphology of mice in each group was observed by naked eye. Uterus weight was weighted. The nerve growth factor( NGF) in endometrium and the receptor P75 expression of mice in each group were detected. Results The uterus surface of mice in the blank group was smooth; no abnormal nodules were found on the surface of the uterus. Adenomyosis nodules could be seen on the surface of the uterus of mice in the model group and the herb group. Uterus weight of mice in the herb group and the model group were higher than that in the blank group( P 0. 05). The deficiency of uterus weight of mice in the herb group and the model group was not significant( P 0. 05). The deficiency of NGF protein expression of mice in each group was not significant( P 0. 05). P75 expression in endometrium of mice in the model group was higher than that in the blank group while P75 expression in endometrium of mice in the herb group was lower than that in the blank group( P 0. 05). Conclusion Yikun Yitongping Keli might relieve AM-ralated dysmenorrheal by reducing the expression of NGF receptor P75 in endometrium.
