不同采摘期蓝萼香茶菜中蓝萼甲素的含量测定及提取工艺的研究

摘要 目的测定不同采摘时期蓝萼香茶菜中蓝萼甲素的含量,考察最适采收期;研究蓝萼香茶菜中蓝萼甲素的提取工艺。方法 HPLC法,色谱柱为Waters Symmetry C_(18)柱(4.6 mm×250 mm,5μm),流动相为甲醇-水(70∶30),检测波长250 nm,流速1.0 ml/min,柱温35℃,进样量10μl;单因素试验法比对不同溶剂、不同提取方式对蓝萼甲素提取率的影响,正交试验法优选蓝萼甲素的乙醇提取工艺。结果 7月份采集的蓝萼香茶菜中蓝萼甲素含量最高;甲醇提取的蓝萼甲素含量最高为1.932 mg/g(n=3),95%乙醇次之为1.896 mg/g(n=3);乙醇提取蓝萼甲素的最佳工艺为95%乙醇加热回流提取2 h,提取1次,固/液比1∶15。结论蓝萼香茶菜最宜采摘时间为7月份;乙醇提取工艺安全、稳定、成本低,适合工业生产。 Objective To determine the content of Glaucocalyxin A in different period of picking and select the optimum harvest time; to study the extraction process of Glaucocalyxin A from Isodon glaucocalyx (Maxim.) Kudo. Methods The separation is carried out on Waters Symmetry Cl8 column (4.6 mm ×250 mm, 5 μm), the mobile phase composed of methanol and water (70 : 30), the detection wavelength is set at 250 nm, the flow rate is 1.0 ml/min, the column temperature is 35℃ and the injection volume is 10 gl; the effect of different solvents and different extraction methods on the extraction rate of Glaucocalyxin A is studied by single factor test, the ethanol extraction process is optimized by orthogonal test. Results In July, the content of Glaucocalyxin A is the highest; Methanol extract have the highest content of Glaucocalyxin A, which is 1.932 mg/g (n = 3), 95 % ethanol is followed by 1.896 mg/g (n = 3); the optimum extraction process of ethanol is 95 % ethanol, refluxing, extracting 2 h, extracting 1 times, solid / liquid is 1 : 15. Conclusion Isodon glaucocalyx (Maxim.) Kudo should be collected in July; the process of ethanol extraction is safer, more stable and less expensive, it is suitable for industrial production.
