摘要
目的:体外评估十株乳杆菌的免疫作用,探讨小鼠离体脾脏淋巴细胞和巨噬细胞在筛选免疫活性乳酸菌中应用的可行性。方法:采用CCK-8法观察不同剂量的十株乳杆菌对小鼠脾淋巴细胞增殖能力以及对小鼠腹腔巨噬细胞能量代谢水平的影响,采用巨噬细胞吞噬中性红法评估不同剂量的十株乳杆菌的对巨噬细胞吞噬作用的影响。结果:十株乳杆菌对脾淋巴细胞增殖活性作用和对巨噬细胞吞噬作用以及对巨噬细胞能量代谢水平的影响均表现出一定的量效关系,其中植物乳杆菌KLDS 1.0318对脾淋巴细胞和巨噬细胞的调节活性最高。结论:作为免疫细胞的小鼠脾淋巴细胞和巨噬细胞离体后培养稳定,可以用于体外实验初步筛选出具备较高潜在免疫活性的乳杆菌,通过体外评估十株乳杆菌对免疫细胞活性的影响,结果表明植物乳杆菌KLDS 1.0318具有较高的免疫调节活性。
Objective:To evaluate the immunomodulation effect of ten Lactobacillus strains in vitro and to explore the application of mouse spleen lymphocytes and macrophage in the selection of lactic acid bacteria with immunological activity. Methods: CCK-8 method was used to observe the effects of different doses of ten strains of Lactobacillus on the proliferation of mouse spleen lymphocytes and the level of energy metabolism, the effects of different doses of ten LactobaciUus strains on phagocytosis of macrophages were evaluated by the method of macrophage phagocytosis of neutral red. Results : The effect of ten strains of Lactobacillus on the proliferation of splenic lymphocytes and the phagocytosis of macrophages as well as the energy metabolism of macrophages showed a dose-effect relationship, among them, Lactobacillus plantarum KLDS 1.0318 had a higher regulation activity on spleen lymphocytes and macrophages. Conclusion : As immune cells, the mouse spleen lymphocytes and abdominal macrophages in vitro can be used to screen the Lactobacillus with potential immune activity. The effect of ten Lactobacillus strains on the activity of immune ceils was evaluated in vitro, and the results showed that Lactobacillus plantarum KLDS 1.0318 had a higher immunomodulatory activity.
