摘要
为了开发与含油量紧密相关、能用于分子辅助高油育种的分子标记,本研究根据与花生含油量相关的位点设计了29对引物,并在7份品种中进行初步筛选。结果显示,有一对引物ID14-3表现出多态性,可以分别扩增出278bp(ID14-3a)和272bp(ID14-3b)的片段。进一步用该引物对171份品种组成的自然群体进行扩增,含ID14-3b的品种平均含油量为48%,含ID14-3a的品种平均含油量为46%。ID14-3b与花生品种的高含油量相关,可以用于花生含油量的分子标记辅助选择育种和高油种质鉴定。
n order to identify molecular markers associated with oil content,which can be used to breeding for high oil content through molecular marker-assisted selection(MAS),29 pairs of primers were designed according to the locus associated with peanut oil content,and screened firstly in 9 peanut varieties.The result showed that a pair of ID14-3 primer amplified polymorphic bands in different above mentioned peanut cultivars,and produced two fragments of 272 bp(ID14-3 a)and 278 bp(ID14-3 b)respectively in different peanut varieties.ID14-3 primer was used to detect 171 varieties.The average oil content of varieties with ID14-3 bwas 48%,while the oil content of varieties with ID14-3 a was 46%.The analysis of nature population showed that the ID14-3 bwas linked to the oil content,and it could be used for MAS breeding and germplasm evaluation for high oil content trait.
