五味子多糖通过Traf3/NF-?B通路降低脂多糖对大鼠肝细胞的炎症反应 被引量:6

Effects of Schizandrae Polysaccharide on Inflammatory Reaction of LPS-induced Rat Hepatocytes via Traf3/NF-?B Signaling Pathway

摘要 目的探讨五味子多糖对脂多糖(LPS)诱导原代大鼠肝细胞炎症反应的影响,并研究潜在机制。方法体外原代培养大鼠肝细胞,并分为空白对照组、LPS诱导组、五味子多糖低、中、高剂量组,CCK-8法检测五味子多糖对大鼠肝细胞增殖的影响,ELISA法检测五味子多糖对大鼠肝细胞分泌IL-6和TNF-α炎症因子的影响,免疫印迹方法检测五味子多糖对Traf3/NF-?B信号通路的影响。结果与LPS诱导组相比,五味子多糖能显著改善LPS对大鼠肝细胞增殖的抑制作用(P<0.05);与LPS诱导组相比,不同剂量五味子多糖均显著降低了大鼠肝细胞分泌的IL-6和TNF-α炎症因子水平(P<0.05);免疫印迹法检测发现,各剂量五味子多糖组相对LPS诱导组细胞中Traf3和p-NF-?B表达水平明显降低(P<0.05)。结论五味子多糖可抑制大鼠肝细胞炎症反应,进而改善炎症对细胞增殖的抑制作用,其作用机制可能与下调Traf3/NF-?B信号通路活化有关。 OBJECTIVE To explore the effects of schizandrae polysaccharide(SP) on inflammatory reaction of lipopolysaccharide(LPS)-induced rat hepatocytes, and study the potential mechanism. METHODS Primary rat hepatocytes were cultured in vitro, and hepatocytes were divided into five groups, including control group, LPS-indueed group, low dose-, medium dose-, and high dose SP groups. The effect of SP on the proliferation of hepatocytes were investigated using CCK-8 assay. ELISA was used to investigate the influence of SP on the secretory levels of inflammatory cytokine (IL-6 and TNF-α). Western blot was used to assess the impacts of SP on the Traf3/NF-κB signaling pathway. RESULTS SP significantly improved the inhibitory effect of LPS-induced hepatocytes proliferation (P〈0.05). Varying concentrations of SP could significantly inhibit the secretion of IL-6 and TNF-α in hepatocytes compared with LPS-induced group (P〈0.05). Western blot results showed that the expression levels of Traf3 and NF-κB in all SP groups significantly decreased compared with LPS-induced group(P〈0.05). CONCLUSION SP can significantly inhibit the inflammatory reaction of rat hepatocytes and its molecular mechanism may involve in down-regulating the activation of Traf3/NF-κB signaline oathwav.
