摘要
目的探讨铜绿假单胞菌(PA)注射液对肺癌细胞增殖、凋亡的影响,并探讨其机制。方法选择肺癌细胞株NCI-H1395和95-D接种于96孔板,分别设对照组和观察1、2、3组,分别加入0、0.25×109/m L、0.5×109/m L、1×109/m L的PA注射液培养24 h。另设对照组、PA组和AG1478组,对照组不加药,PA组加入1×109/m L的PA,AG1478组加入1×109/m L的PA和10-7mol/L的表皮生长因子受体抑制剂AG1478。采用MTT法观察各组细胞增殖情况;Annexin V/PI双染试验检测各组细胞凋亡率。结果随着PA浓度的升高,肺癌NCI-H1395和95-D细胞OD570不断降低,细胞凋亡率不断升高(P均<0.05)。与对照组相比,PA组细胞OD570降低(P<0.05),细胞凋亡率升高(P<0.05);与PA组相比,AG1478组细胞OD570升高,细胞凋亡率降低(P均<0.05)。结论 PA注射液通过EGFR信号通路抑制肺癌细胞NCI-H1395和95-D增殖,促进其凋亡。
Objective To explore the effects of pseudomonas aeruginosa( PA) injection on the cell proliferation and apoptosis of lung cancer cells,and to explore its mechanism. Methods NCI-H1395 and 95-D of lung cancer cell lines were selected and then were inoculated in 96-well plate. We divided them into the control group and observation groups 1,2,and 3,which were separately added with 0,0. 25 × 109,0. 5 × 109,and 1 × 109/m L PA injection for 24 h. In addition,the control group,PA group,and AG1478 group were set. The drug was not added into the control group,1 × 109 PA was added into the PA group,and 1 × 109/m L PA and 10-7 mol/L EGFR inhibitor AG1478 were added into the AG1478 group. The proliferation of cells was observed by MTT. The apoptosis rate of each group was detected by the Annexin V/PI double dye test. Results With the increase of PA concentration,the OD570 of NCI-H1395 and 95-D cells was decreasing,and the cell apoptosis rate increased( all P 〈0. 05). Compared with the control group,the OD570 of PA group decreased( P〈 0. 05),while the apoptosis rate increased( P〈 0. 05). Compared with the PA group,the OD570 of AG1478 increased,and the apoptosis rate decreased( both P 〈0. 05). Conclusion PA injection can inhibit the proliferation of NCIH1395 and 95-D of lung cancer cells and promote apoptosis through EGFR signaling pathway.
