摘要
目的比较CMU/1和CMU/2近交系长爪沙鼠脑底动脉Willis环缺失比率,检测血液生理生化指标,分析比较二者差异,为其实验应用提供参考数据。方法采用Beckman全自动生化分析仪(CX5)和日本光电全自动血细胞分析仪(MEK-7222K)对17项血液生化指标:总蛋白(TP)、白蛋白(ALB)、总胆固醇(CHOL)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDLD)、尿素氮(BUN)、肌酐(CRE)、尿酸(URIC)、总胆红素(TBIL)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、乳酸脱氢酶(LD)、肌酸激酶(CK)、淀粉酶(AMY)、镁(Mg)以及葡萄糖(GLU)和22项血液生理指标:白细胞(WBC)、淋巴细胞(LYM)、中性粒细胞(NEUT),单核细胞(MON)、嗜酸性粒细胞(EOS)、嗜碱性粒细胞(BAS)、淋巴细胞百分比(LYM%)、中性粒细胞百分比(NEUT%)、单核细胞百分比(MON%)、嗜酸性粒细胞百分比(EOS%)、嗜碱性粒细胞百分比(BAS%)、红细胞(RBC)、平均红细胞血红蛋白含量(MCH)、红细胞体积分布宽度(RDW),血红蛋白(HGB)、平均红细胞血红蛋白浓度(MCHC)、红细胞压积(HCT)、红细胞平均体积(MCV)、血小板(PLT)、血小板压积(PCT)、血小板平均体积(MPV)、血小板体积分布宽度(PDW)进行测定。应用SPSS23.0软件进行独立样本t检验的统计分析。结果CMU/1和CMU/2近交系长爪沙鼠脑底动脉Willis环缺失比例均为76%以上;血液生理生化的12项指标:GLU、CRE、TG、CHOL、PLT、MPV、BAS、LYM%、PDW、HCT、MCV、MCH雌雄差异显著;CMU/1和CMU/2雌性ALT、PLT、PDW差异显著,雄性的HDLD、MCH差异显著;品系间3项指标ALP、ALT、AST差异显著。结论脑缺血模型长爪沙鼠近交系的基因型对血液生化和生理指标有一定的影响。
Objective To compare the ratio of the incomplete circle of Willis patterns and detect the blood biochemical and physiological indexes of 2 ischemia-prone inbred gerbil lines CMU/1 and CMU/2, and to analyze the differences between 2 strains, provide reference data for gerbils experimental application. Methods Beckman automatic biochemical analyzer (CX5) and Japan photoelectric automatic blood cell analyzer (MEK-7222K) were used to determine 17 blood biochemical parameters and 22 physiological parameters. SPSS 23.0 software was used for statistical analysis by independent samples T test. Results The ratios of incomplete circle of Willis patterns in both ischemia-prone inbred gerbil lines CMU/1 and CMU/2 were more than 76%. The 12 parameters of blood physiology and biochemistry, glucose, creatinine, triglycerides, cholesterol, platelet, mean platelet volume, basophilic granulocyte, lymphocyte%, platelet distribution width, hematocrit, mean corpuscular volume, mean Corpuscular hemoglobin, were significantly different between male and female. CMU/1 and CMU/2 were significantly different in female alanine transaminase, platelet and platelet distribution width, and high-density lipopro- rein cholesterol and mean corpuscular hemoglobin in males were significantly different. There were significant differences in alkaline phosphatase, alanine transaminase and aspartate transaminase between strains. Conclusion The genotypes of ischemia-prone inbred gerbil have some impact on blood biochemical and physiological parameters.
