桑蒺颗粒中陈皮、枳壳挥发油的提取及β-环糊精包合工艺 被引量:6

摘要 目的:优选桑蒺颗粒中陈皮、枳壳挥发油的最佳提取及包合工艺。方法:采用正交试验法,以挥发油提取量为评价指标,优化加水量、浸泡时间和提取时间3个因素,优选挥发油提取工艺;以挥发油包合率和包合物收率为评价指标,优化挥发油︰β-环糊精(β-CD)、包合温度和包合时间,优选挥发油包合工艺;采用显微镜、薄层色谱及加速试验对所得包合物进行表征。结果:桑蒺颗粒中陈皮、枳壳挥发油的最佳提取工艺为加8倍量水,浸泡0.5 h,提取6 h;最佳包合工艺为挥发油与β-CD比例1∶10,在40℃下包合0.5 h;3项表征试验均证实挥发油已包合于β-CD内。结论:优选的桑蒺颗粒中陈皮、枳壳挥发油的最佳提取及包合工艺稳定可行。 OBJECTIVE To optimize extraction technology of volatile oils fromCitri Reticulatae Pericarpium and Aurantii Fructus and theβ-cyclodextrin inclusion process for Sangji Granules.METHODS The orthogonal tests were designed to optimize extraction technology,with the yield of the volatile oil as indexes,the ratio of water to herbal medicine material,the time of dip in water and the time of distillation were used as the main factors.With yield of inclusion complex and inclusion rate as comprehensive indexes,the ratio ofβ-cyclodextrin(β-CD)to volatile oil,inclusion temperature,inclusion time as factors,the inclusion process of volatile oil from Sangji Granules was optimized.The inclusion complex was characterized with microscopic imaging,TLC and accelerated test.RESULTS The best extraction technology was as follows:the time of dip in water was 0.5 h,the ratio of water to herbal medicine materials 8:1,the time of distillation 6 h.The optimum inclusion process was as follows:ratio of volatile oil toβ-CD was 1∶10,inclusion temperature 40 ℃,mixing time 0.5 h.The 3 characterization tests indicated that volatile oil had been included inβ-CD.CONCLUSION The optimized extraction technology andβ-cyclodextrin inclusion process are feasible and stable.
