摘要
用组织培养法将人正常肾上腺皮质及原发性醛固酮增多症腺瘤(aldosterone-pro-ducing adenoma,APA)组织,在常规细胞培养条件下培养,以醛固酮分泌作为组织存活指标。用α-人心钠素(α-human atrial natriuretic facfor,α-hANF)处理体外培养组织(正常组织用量为10^(-8)mol/L,APA组织为3×10~8mol/L),结果α-hANF作用后30min、2、4及24h,两种组织均有反应,正常组织表现为醛固酮分泌增加;APA则表现为轻微抑制。
Human adrenal gland and aldosterone-producing adenoma (APA)obtained from adrenalectomy could be maintained in vitro for one to two months. The explants continued to secrete aldosterone for the duration of culture. This method was superior to monolayer cell culture. The influence of α-human atrial natriu-retic factor (α-hANF) was investigated by treating cultures of normal adrenal cortex and APA with α-hANF (10-8 mol/L and 3 × 10-8 mol/L respectively). We measured aldosterone in culture media collected at 2h before a-hANF treatment and 0.5.2,4 and 24h after. It was found that a-hANF stimulated aldosterone secretion of normal adrenal tissue but slightly inhibited that of APA.
