高效液相色谱法测定升血小板胶囊中靛玉红的含量

Content Determination of Indirubin in Shengxuexiaoban Capsules by High Performance Liquid Chromatography

目的:建立升血小板胶囊中靛玉红的含量测定方法。方法:采用高效液相色谱法测定升血小板胶囊中靛玉红的含量。色谱柱为依利特ODS-C_(18)柱(150 mm×4.6 mm,5μm);流动相为甲醇-1%甲酸溶液(V:V=30:70);柱温为30℃;流速为0.8 ml/min;检测波长为289 nm。结果:靛玉红质量浓度在0.20~5.08μg/ml范围内线性关系良好(r=0.999 9);精密度、稳定性和重复性试验的RSD均<2%;样品加样回收率为98.97%~100.92%,RSD均<2%。结论:本方法简便、灵敏及准确,专属性强,可作为升血小板胶囊的含量测定方法。

OBJECTIVE ： To establish method for content determination of indirubin in Shengxuexiaoban capsules. METHODS： High performance liquid chromatography （HPLC） was adopted to detect the content of indirubin in Shengxuexiaoban capsules. The ODS-C^s （150 mm x4. 6 ram, 5 txm） was set as chromatographic column; mobile phase was methanol-1% of formic acid （ V： V = 30： 70） ; column temperature was 30 ~C ; flow rate was 0. 8 ml/min ; detection wavelength was 289 nm. RESULTS ： The linear relation remained well （ r = 0. 999 9 ） when the mass concentration of indirubin stayed within 0.20-5.08 txg/ml ; all the relative standard deviations （RSD） of precision degree, stability and repetitive test were 〈 2% ; the recovery rate of sample addition was 98.97% -100.92%, with RSD 〈 2%. CONCLUSIONS： This HPLC method is convenient, sensitive and accurate, with strong specificity, which can be served as the method for content determination of Shengxuexiaoban capsules.
