海洛因依赖大鼠颌下腺IL-1、TNF表达的改变 被引量:3

摘要 目的观察白细胞介素-1(interleukin-1,IL-1)、肿瘤坏死因子(Tumor Necrosis Factor,TNF)在大鼠颌下腺的表达情况以及在海洛因依赖时表达的改变。方法成年雄性SD大鼠,按配对原则随机分为正常对照组(normal control group,NCG)、盐水对照组(saline control group,SCG)、海洛因依赖组(heroin dependence group,HDG)。于大鼠腹部皮下注射海洛因药液,2次/d(上午8时,下午3时),至成瘾后取颌下腺组织,应用免疫组织化学Envision法及图像分析法进行研究。结果 1.在正常大鼠,IL-1、TNF阳性细胞分布于颌下腺导管系统,其中以颗粒曲管(Granular Convoluted Tubule,GCT)细胞为主,腺泡细胞为阴性;2.与正常大鼠相比,盐水对照组大鼠颌下腺IL-1、TNF阳性细胞的数量及平均灰度值均无明显改变(P>0.05);3.海洛因依赖组大鼠颌下腺IL-1、TNF阳性细胞的分布未见明显变化,但细胞数量增加(P<0.05);4.图像分析结果显示IL-1、TNF阳性细胞的平均灰度值低于正常及盐水对照组大鼠(P<0.05)。结论海洛因依赖期间大鼠颌下腺IL-1、TNF的合成和分泌增加,海洛因对颌下腺神经内分泌功能有明显影响。 Objective To investigate the location features and changes of IL-1 and TNF in submandibular gland of heroin dependence rats. Method Male rats were divided into normal control group (NCG) , saline control group (SCG) and heroin dependence group (HDG). To establish the heroin dependence model of rats by abdominal subcutaneous injection with heroin( Bid, 8:00AM and 14:00PM) and excise the submandibular glands to perform immunohistochemical envision method and image analysis to detect the expression of IL-1 and TNF. Result 1. The location of IL-1 and TNF positive cells were in the duct system of submandibular gland, moreover, it was granular convoluted tubule (GCT) mainly. But the acinus cells were negative. 2. As compared with the normal control group rats, the numbers and the mean grey values of IL-1 and TNF positive cells did not change obviously (P 〉 0.05). 3. The location of IL-1 and TNF positive cells did not change obviously, the cell numbers increased obviously yet in herion dependence group rats (P 〈 0.05). 4. The result of image analysis showed that the mean grey values of IL-1 and TNF positive cells decreased in heiron dependence group rats (P 〈 0.05). Conclusion The synthesis and secretion of IL-1 and TNF increased in heroin dependence rats. Heroin affected the neuroendocrine function of submandibular gland obviously.
