新型树脂对内毒素吸附的体外研究 被引量:1

摘要 目的探讨新型吸附树脂(EHC-I)对内毒素(ET)吸附性能的研究。方法采用静态吸附实验检测水及血浆内毒素下降率,将内毒素分别加到水及血浆中配制一定浓度的内毒素溶液,树脂与血浆及树脂与水按比例混合,置37℃恒温水浴按一定速度连续震荡2 h,采用动态浊度法在630 nm处测定震荡前、后内毒素水平,然后计算吸附率。同时做溶血实验以检测树脂的生物相容性。结果内毒素水溶液的下降率为94.86%,内毒素血浆溶液的下降率为89.06%,溶血实验显示溶血率为2.86%。结论新型树脂对内毒素有明显吸附效果,溶血率符合要求,为研制适用于临床的高效清除内毒素的血液灌流器提供了依据。 Objective To study the efficacy of new type resin (EHC-I) for the adsorption of endotoxin in vitro. Method The descent rate of endotoxin in water and plasma were measured by static adsorption experiment. Endotoxin were added into water and plasma, then they proportionally-mixed with resin, mixtures were putted into constant temperature water bath oscillator at 37 ℃ , and had been vibrated for 2 h. Before and after the experiment, endotoxin were detected at 630 nm by kinetic turbidimetric assay, then to calculate the adsorption rate, meanwhile hemolysis test was performed to detect the biocompatibility of the resin. Result The decreasing rate of endotoxin in aqueous solution was 94.86% , that of in plasma was 89.06% , hemolytic rate was 2.86%. Conclution The new type resin has a significant adsorption effect on endotoxin, hemolytic rate meets the requirements, which provides a basis for the development of hemoperfusion devices that suitable for clinical efficient clearance of endotoxin.
