胃癌组织中VGLL4、YAP蛋白表达与胃癌患者预后的关系

Relationship between the expression of VGLL4 and YAP protein in gastric cancer tissue and prognosis of patients with gastric cancer

目的检测胃癌组织及癌旁组织中VGLL4、YAP蛋白的表达情况,探讨其在胃癌诊治中的临床意义。方法采用免疫组织化学SP法检测46例胃癌组织及癌旁组织中VGLL4、YAP蛋白的表达情况,分析其表达与胃癌患者预后的相关性。结果胃癌组织中VGLL4、YAP蛋白的阳性表达率分别为39.1%(18/46),76.1%(35/46),癌旁组织中的阳性表达率分别为91.3%(42/46)、10.9%(5/46),差异有统计学意义(P<0.05)。VGLL4和YAP蛋白的表达均与胃癌组织学分级、临床分期、淋巴结转移、侵袭性具有相关性(P<0.05),且与患者预后相关。YAP阳性表达高水平胃癌患者术后中位生存时间[(19.000±0.557)个月]明显短于YAP阳性表达低水平胃癌患者[(36.000±2.942)个月],VGLL4阳性表达低水平胃癌患者术后中位生存时间[(18.000±1.503)个月]明显短于VGLL4阳性表达高水平胃癌患者[(26.000±2.863)个月],YAP和VGLL4表达水平均与胃癌患者预后明显相关,差异有统计学意义(P<0.05)。结论VGLL4在胃癌组织中较癌旁组织表达下降,且与肿瘤发展和恶性程度呈负相关,是一种潜在的抑癌蛋白。YAP在胃癌组织中较癌旁组织表达增高,与肿瘤发展和恶性程度呈正相关,且两者均与患者预后相关。

Objective To detect the expression of vestigial-like family member 4（VGLL4） and yes-associatedprotein（YAP） in gastric cancer tissue and paracancerous tissue, and to analyze its clinical significance in the diagnosis andtreatment of gastric cancer. Methods The expression of VGLL4 and YAP protein in 46 cases of gastric carcinoma andparacancerous tissue were detected by SP immunohistochemical method, and the correlation between the expression andprognosis of patients was analyzed. Results The positive expression rates of VGLL4 and YAP protein were respectively39.1%（18/46） and 76.1%（35/46） in gastric cancer tissues versus 91.3%（42/46） and 10.9%（5/46） in paracancerous tissues（P〈0.05）. The expression of VGLL4 and YAP protein were correlated with the histological grade, clinical stage, lymph nodemetastasis and invasion（P〈0.05）, and correlated with the prognosis. The median survival of patients with high level of YAPpositive expression was（19.000±0.557） months, which was significantly shorter than（36.000±2.942） months of the patientswith low levels of YAP positive expression（P〈0.05）; the median survival of patients with low level of VGLL4 positive expres-sion was（18.000±1.503） months, which was significantly shorter than（26.000±2.863） months of the patients with high levels ofVGLL4 positive expression（P〈0.05）. Conclusion Compared with the adjacent tissue, the expression of VGLL4 in gastric can-cer tissue shows a significant downward trend, and the degree of tumor development and malignancy is negatively correlatedwith the expression of VGLL4, as a potential anti-cancer protein. YAP is a proto-oncogene, which is highly expressed in gastriccancer tissues, and positively correlated with tumor progression and malignancy. The expression of VGLL4 and YAP protein iscorrelated with the prognosis of gastric cancer.
