星虫多糖对亚急性辐射损伤小鼠造血系统的保护作用研究 被引量:5

Study on hematopoietic radioprotection effect of Sipunculus nudus L. polysaccharide on sub-acute radiation-injured mice

摘要 目的研究星虫多糖(Sipunculus nudus L.polysaccharide,SNP)对亚急性辐射损伤小鼠造血系统的保护作用。方法选用雄性BALB/c,采用^(137)Csγ射线对小鼠进行1次性全身照射(总剂量为4.0Gy),建立亚急性辐射损伤模型。照射前连续灌胃给药7d,照射后继续给药14d。用细胞分析仪测定外周血象,流式细胞仪检测骨髓造血干细胞含量与细胞凋亡率。照射后24h取小鼠股骨进行HE染色观察骨髓病理变化。结果经^(137)Csγ射线照射后,与辐照模型组相比,照后第10天,90mg/kg SNP小鼠外周血象红细胞数量显著提高(P<0.05),SNP 3个剂量组血小板数量均显著提高(P<0.05);照后第14天,270mg/kg SNP组小鼠脾脏指数与30mg/kg SNP组小鼠睾丸指数显著提高(P<0.05),30mg/kg SNP组骨髓造血干细胞数量显著提高(P<0.05),SNP各给药组骨髓细胞凋亡率显著下降(P<0.01),骨髓组织中有核细胞数量显著增多。结论SNP对辐射损伤小鼠的造血系统具有较好的保护效果。 Objective To study the hematopoietic radioprotection of SipunculusnudusL.polysaccharide on sub-acute radiation-injured mice.Methods To establish sub-acute radiation-injured rat model,male BALB/c mice were irradiated by ^(137)Csγ-rays at the dose rate of 0.75 Gy/min,and the accumulated dose was 4.0 Gy.The mice were administered with SNP intragastrically for 7 days before irradiation and for 14 days after irradiation.RBCs,WBCs and PLTs in peripheral blood were counted by cytoanalyze.The organ index was detected,and the content of hematopoietic stem cells and apoptosis rate of cells in bone marrow were measured by flow cytometry.The bone marrow histology pathology was observed by HE staining on 24 hafter irradiation.Results Compared with radiation control group,on 10th day after irradiation,RBCs in peripheral blood of 90 mg/kg SNP group and PLTs in three SNP treated groups were increased significantly(P0.05).On 14 thday after irradiation,the spleen index of 270 mg/kg SNP group and testis index of 270 mg/kg SNP group were increased significantly(P0.05).The hematopoietic stem cells in bone marrow of 30 mg/kg SNP group were increased significantly(P0.05),and the apoptosis rates of three SNP treated groups were decreased remarkably(P0.01).HE staining showed that treatment with SNP increased the karyocyte amounts in bone marrow.Conclusion These findings suggested that SNP possessed marked hematopoietic protection on damage in mice induced by137 Csγ-irradiation.
