山茱萸新苷抑制EAE模型大鼠中枢神经系统免疫细胞浸润及VCAM-1表达的研究 被引量:8

摘要 目的明确山茱萸新苷是否可以抑制实验性自身免疫性脑脊髓炎(EAE)模型大鼠中枢神经系统免疫细胞浸润及血管细胞黏附分子-1(VCAM-1)表达。方法 Lewis大鼠32只,随机分为正常对照组、模型组、强的松组以及山茱萸新苷组,各8只。除正常对照组外,均制作EAE模型。每天评定各组大鼠神经功能缺损症状评分,当症状达到高峰期时处死所有大鼠,比较各组大鼠脊髓炎性病灶数以及VCAM-1 mRNA、VCAM-1蛋白表达。结果正常对照组、模型组、强的松组以及山茱萸新苷组大鼠第14d神经功能缺损症状评分分别为0分、(3.83±0.98)分、(0.67±0.82)分、(1.67±1.75)分,脊髓炎性病灶个数分别为0个、(8.75±2.23)个、(2.33±1.03)个、(4.83±1.47)个,VCAM-1mRNA的相对表达量分别为(5.602±0.761)、(37.762±4.560)、(17.143±1.521)、(28.042±3.147),VCAM-1蛋白相对表达量分别为(7.958±2.265)、(22.433±1.829)、(15.725±1.338)、(18.625±0.874)。与模型组比较,山茱萸新苷组大鼠神经功能缺损症状评分较低,脊髓炎性病灶个数较少,VCAM-1mRNA相对表达量及VCAM-1蛋白的相对表达量较低(P<0.05,P<0.01)。结论山茱萸新苷可以改善EAE模型大鼠神经功能缺损症状,抑制免疫细胞浸润EAE模型大鼠中枢神经系统及中枢神经系统VCAM-1 mRNA及蛋白质表达。 Objective To investigate the effect of cornuside on the invasion of immune cells and the expression of VCAM-1 in the central nervous system of experimental autoimmune encephalomyelitis(EAE) rats. Methods Thirty-two Lewis rats were randomly divided into normal control group, model group, prednisone group and cornuside group, with 8 rats in each group. Except for normal control group, EAE model was prepared in other groups.The neurological deficit symptoms of rats were daily observed. All rats were sacrificed when the neurologic deficits reached peak stage. HE staining was used to observe the severity of the invasion of immune cells, and real time quantitative PCR(RT-PCR) was used to determine the expression of VCAM-1 mRNA and Western Blotting to detect the expression of VCAM-1 protein. Results On the day 14, the score of neurological deficit symptoms of rats in normal control, model, prednisone, and cornuside groups were 0, 3.83 ±0.98, 0.67 ±0.82, 1.67 ±1.75, the number of lesions stained by HE were 0, 8.75±2.23, 2.33±1.03, 4.83±1.47, the relative expression of VCAM-1 mRNA were5.602 ±0.761, 37.762 ±4.560, 17.143 ±1.521, 28.042 ±3.147, and the relative expression of VCAM-1 protein were7.958±2.265, 22.433±1.829, 15.725±1.338, 18.625±0874, respectively. Compared with model group, cornuside group had lower score of neurological deficit symptoms, less number of lesions, lower relative expression of VCAM-1 mRNA and protein in the central nervous system(P〈0.05 or 0.01). Conclusion Cornuside can alleviate neurological deficit symptoms, suppress invasion of immune cells, and inhibit the expression of VCAM-1 mRNA and protein in the central nervous system in rats with EAE.
