摘要
目的观察竹节参水、醇提取物对慢性酒精性肝损伤模型小鼠血生化指标及炎症因子水平的干预作用。方法 ICR小鼠40只,随机分为正常组、模型组、易善复组、竹节参醇提取物(PJEE)组、竹节参水提取物(PJWE)组,各8只。采用灌胃一定浓度乙醇溶液复制慢性酒精性肝损伤小鼠模型,同时予以竹节参水、醇提取物(1.35g/kg)干预;与第2、4、6周全自动生化仪检测血清肝功能指标、血脂水平;第4、6周酶联免疫吸附法(ELISA)检测血清白介素-6(IL-6)、白介素-1β(IL-1β)水平。结果与正常组比较,自第2周起,模型组小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性随着造模时间的延长而显著增加(P<0.05,P<0.01),血清总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)水平随着造模时间的延长而呈逐渐降低趋势(P<0.05),第4、6周血清IL-6、IL-1β水平均显著上升(P<0.01)。与模型组比较,竹节参醇提取物组血清ALT、AST、TG显著降低(P<0.05),竹节参水、醇提取物组血清IL-6、IL-1β水平组均显著降低(P<0.01),其中第4周以水提取物较为显著,第6周以醇提取物较为明显。结论竹节参水、醇提取物均具有一定的抗慢性酒精性肝损伤作用,但在作用侧重点、作用时间和强度间存在一定差异。
Objective To study the effect of panax japonicus water or ethonal extractions on blood biochemical indexes and inflammatory cytokines in mice with alcoholic liver injury(ALI). Methods ALI model was established in ICR mice by gavage with gradient alcoholic solution. Mice in experiment groups were orally administered with water panax japonicus extraction(PJWE) or ethanol panax japonicus extraction(PJEE) for 6 weeks. Blood were collected at 2, 4 and 6 week to determine serum alanine aminotransferase(ALT), aspartate aminotransferase(AST), total cholesterol(TC), triglyceride(TG), high-density lipoprotein cholesterol(HDL-C) with automatic biochemical analyser and serum IL-6, IL-1β by ELISA according to the specification of the kits. Results From 2 weeks, serum ALT and AST activities in ALI mice were increased with the prolong of the ethanol intake(P〈0.05, P〈0.01), while serum TC and HDL-C levels were decreased on the contrary(P〈0.05); high levels of serum IL-6 and IL-1β were observed at 4 week and 6 week(P〈0.01). At 4 week PJEE significantly lowered serum ALT and AST activities(P〈0.05), but no obviously changes were found in serum lipid levels. Both PJWE and PJEE reduced serum IL-6 and IL-1β levels, and at 4 w the effects of PJWE was stronger and at 6 week was PJEE. Conclusion Both PJWE and PJEE posses an effect on anti ALI, but there are some differences in effect emphasis, working time and action intensity between them.
