人X染色体α卫星DNA的定位及应用初探

摘要 应用人X染色体α卫星DNA探针对正常男女及45,XO/47,XXX个体外周血淋巴细胞的原位杂交研究表明,在R显带的中期分裂相中,绝大部分杂交颗粒位于X染色体着丝粒区,即p11→q11区,在间期核内,则呈现与X染色体数目明显相关的银颗粒簇,其中44%位于核边缘区。本实验显示α卫星DNA的染色体区域定位及在具有不同X染色体数目间期核内分布的高度特异性。 In situ hybridization of human A chromosome-specitic alphoid satellite DNA to human lymphocyte metaphase and interphase nuclei was carried out. The results showed that the probe hybridized with high specificity to the centro-meric region at p11→qll of the X chromosome. The number of silver grain clusters in the interphase nuclei was correlated with that of X chromosomes in the cells. 44% of the clusters were located near the nuclear membrane, the usual location of X chromatin (Barr body) . The hybridized chromosomes were also R-banded with BrdU treatment, and the significance of this work was briefly discussed.
