摘要
目的建立高效液相色谱法(high performance liquid chromatography,HPLC)检测保健食品中β-葡聚糖含量。方法称取样品适量,在酸性条件下于135℃用微波消解仪提取15 min,取出水解液,用氢氧化钠溶液调节pH至6.5±0.2,用水定容至100 mL容量瓶中,摇匀后过滤。采用高效液相色谱法检测,根据葡萄糖标准品的色谱峰面积进行定量。结果葡萄糖浓度在0.01~0.16 mg/mL的范围内与峰面积的线性良好,相关系数为0.9998。3水平不同浓度葡萄糖标准品添加的回收率为97.4%~98.6%,相对标准偏差(relative standard deviation,RSD)为0.4%~0.9%。结论该方法操作简便、准确、重现性好,适用于保健食品中β-葡聚糖含量的快速测定。
Objective To establish a method for the determination of β-glucan in health food by high performance liquid chromatography (HPLC). Method The samples were under acidic conditions at 135℃ for microwave extraction of 15 min, and the digestion solution was removed. Sodium hydroxide solution was used to adjust pH to 6.54-0.2. The volume was determined to 100 mL, shaking well and filtering. HPLC was used for detection, and it was quantitated according to the peak area of the glucose standard. Results Glucose concentration was good linear with peak area in the range of 0.01-0.16 mg/mL, with the correlation coefficient of 0.9998. The recoveries were 97.4%-98.6%, with relative standard deviation (RSD) of 0.4%-0.9%. Conclusion The method is simple, accurate and reproducible, which is suitable for the rapid determination offl-glucan in health food.
