发育及DNA损伤反应调节基因1在胎儿生长受限胎盘中的表达及其意义

摘要 目的探讨发育及DNA损伤反应调节基因1(REDD1)在胎儿生长受限(FGR)胎盘中的表达情况。方法收集2011年6月-2012年12月在上海市长宁区妇幼保健院剖宫产或经阴道分娩的足月单胎FGR和正常胎盘各15例,分别作为研究组和对照组,采用免疫组织化学染色方法检测REDD1在胎盘组织中的表达;采用实时荧光定量PCR和Western blotting方法检测REDD1在正常和FGR胎盘组织中mRNA和蛋白水平的变化。结果 REDD1蛋白主要表达于胎盘滋养细胞胞质中,FGR胎盘组织中REDD1 mRNA和蛋白水平均显著高于正常对照组,差异有统计学意义(P<0.05)。结论发育及DNA损伤反应调节基因1可能参与FGR的发生机制。 Objective To explore the expression of regulated in development and DNA damage response 1( REDD1) in placenta of patients with fetal growth restriction( FGR). Methods Fifteen patients( full-term pregnancy,singleton pregnancy) with FGR and fifteen cases( full-term pregnancy,singleton pregnancy) without FGR undergoing cesarean section or vaginal delivery in Maternal and Child Health Care Hospital of Changning District in Shanghai city from June 2011 to December 2012 were collected,then 30 placental samples were obtained as study group and control group,respectively. Immunohistochemical staining was used to detect the expression of REDD1 in placenta. Real-time fluorescent quantitative PCR and Western blotting were used to detect the changes of REDD1 mRNA and protein levels in the two groups. Results REDD1 protein was mainly expressed in cytoplasm of trophoblasts. REDD1 mRNA and protein levels in study group were statistically significantly higher than those in control group( P0. 05). Conclusion REDD1 may play an important role in pathogenesis of FGR.
