摘要
采用改进的细胞移动测量方法,观察巨噬细胞分泌因子对动脉平滑肌细胞移动的直接刺激作用。传代接种的培养细胞以平血中琼脂糖层中心孔为界贴壁形成圆形集落,经照射后分组培养,定期测量细胞集落向外扩展的面积。结果表明,在巨噬细胞条件培养液中,平滑肌细胞每天向外移动约10.6mm^2,在无血小板及全血血清培养液中,分别为5.7及8.6mm^2。实验结果提示巨噬细胞的分泌产物具有直接促进动脉平滑肌细胞移动的化学促动作用。
Stimulation by macrophage secretory products of the migration of arterial smooth muscle cells was studied in vitro with a modified system. Round cell colonies were formed by plating subcultured smooth muscle cells into the middle wells of agarose gels. Following irradiation (1500 rads) and refeeding with test medium, cell migration was observed by measuring the increase in surface area of call colonies at specific time intervals. The cell colonies expanded under the agarose gels with an increment of approximately 10. 6mm^2 per day when exposed to macrophage conditioned medium, and the average increase in sur- face area of the colonies was approximately 5. 7 and 8. 6mm^2 per day when ex- posed to platelet--deficient serum and whole blood serum, respectively. The re- sults indicate that macrophages may contribute to atheromatous plaque forma- tion by stimulating smooth muscle cell migration into the intima in the case of arterial damage due to hyerlipemia or other causes.
