摘要 目的:优化龙眼叶中总黄酮的最佳提取工艺并测定其含量。方法:首先进行单因素实验,然后根据单因素实验的结果确定Box-Behnken响应面实验设计的因素和水平,考察乙醇浓度(A)、乙醇用量(B)、提取时间(C)三个影响因素对龙眼叶总黄酮的提取工艺的影响。结果:经过实验得到的最佳提取工艺为:乙醇浓度53%,乙醇用量57 m L,回流时间83 min,此条件下总黄酮的提取率为15.479%,与预测值(15.482%)相差甚微。结论:本方法不仅龙眼叶总黄酮提取率较高,而且整个实验操作过程较为简便可行,可为龙眼叶药材的进一步研究提供参考。 Objective : To optimize the extraction process of total flavonoids from the leaf of Dimocarpus Longan and determine its content. Methods : The single factor experiments first, and then according to the results to determine the factors and levels of Box - Behnken response surface experiment design, finally, to investigate effects of ethanol concentration ( A ), dosage of ethanol ( B ) and extraction time (C) on comprehensive score of extracting amounts of total flavonoids from the leaf of Dimocarpus Longan. Re- sults :Through experiments we got the best process were as follows:the ethanol concentration was 53% ,the dosage of ethanol was 57 mL and the extraction time was 83 min. The extraction rate of total flavonoids was 15. 479% and the actual value compared with predicted value had little difference. Conclusion : This method not only has high extraction rate of total flavonoids from the leaf of Dimocarpus Longan but also is simple and feasible, providing a theoretical reference for the further study of the leaf of Dimocarpus Longan.
