摘要
目的:通过观察茵栀黄注射液和生脉注射液佐剂组与未联用佐剂组在过敏反应发生时的敏感指标的含量变化,研究此类佐剂提高BN大鼠过敏反应动物模型稳定性和灵敏度的能力。方法:将弗氏完全佐剂与双黄连注射液、生脉注射液联用,对BN大鼠进行致敏和激发,比较联用弗氏佐剂组与未加弗氏佐剂组动物在过敏反应发生率、过敏反应发生程度;激发前后血浆中组胺、类胰蛋白酶、β-氨基己糖苷酶、Ig E含量及增长率;被动皮肤过敏试验大鼠皮下是否出现蓝斑以及蓝斑面积大小。结果:(1)联用佐剂组过敏反应的发生率以及过敏反应的发生程度均高于未加佐剂组;(2)茵栀黄注射液组激发前后血浆中组胺、类胰蛋白酶、β-氨基己糖苷酶、Ig E的含量与生理盐水组比较无显著性差异;联用佐剂后血浆中Ig E、组胺以及β-氨基己糖苷酶的含量均升高,与生理盐水组相比具有显著性的差异,激发前后四种指标的变化率均高于茵栀黄注射液未加佐剂组;生脉注射液组激发前后血浆中四种指标的含量与生理盐水组比较无显著性差异;联用佐剂后血浆中Ig E、β-氨基己糖苷酶的含量均升高,与生理盐水组相比具有显著性的差异,激发前后四种指标的变化率均高于生脉注射液未加佐剂组;(3)联用佐剂组的被动皮肤过敏反应的阳性率均高于未加佐剂组。结论:弗氏完全佐剂可提高过敏反应动物模型的敏感性,有利于中药注射剂安全性中过敏反应的再评价。
Objective : Through the observation of the changes of sensitive index of allergic reaction, adjuvants are used to im- prove sensitivity of BN rats animal models for evaluating anaphylactic reaction. Methods. Yinzhihuang Injection and Shengmai In- jection were mixtured with Freund complete adjuvant ( 1 : 1 ) respectively and sensitized samples in each group. Incidence rates and degrees of allergenic reaction were observed. The difference between adjuvant groups and normal injection groups was com- pared by tryptase, histamine, 13 - hexosaminidase, IgE release and PCA test. Results ~ ( 1 ) The allergic rates of adjuvant groups were higher than those of the normal injection groups and the allergic degree was more serious than that of normal injection groups. (2) IgE, histamine, tryptase, [3 - hexosaminidase concentration of normal injection group with the corresponding normal saline group had no significant differences. Compared with normal saline group, the levels of IgE, histamine and ~3 - Hex in the plasma of Yinzhihuang Injection adjuvant groups increased significantly and the levels of tryptase, [3 - hexosaminidase in the plasma of Shengmai Injection adjuvant groups increased significantly. The release rates of IgE, histamine, tryptase and [3 - hexosaminidase in the plasma of adjuvant groups were higher than that in normal injection groups. ( 3 ) PCA test by using sera from adjuvant group af- ter irritations showed the strong positive result characterized as large amount of subcutaneous effusions of Evans blue in SD rats and the rates of positive response were higher than that of normal injection groups. Conclusion : According to the analysis above,Freund complete adjuvant can be used to improve the sensitivity of the animal model for evaluating anaphylactic reaction and eval- uating the safety of traditional Chinese medicine injection.
