大孔树脂分离纯化伊犁野核桃叶总黄酮的工艺研究 被引量:2

摘要 以伊犁野核桃叶总黄酮含量为响应值,研究大孔树脂分离纯化伊犁野核桃叶总黄酮工艺.比较9种树脂对伊犁野核桃叶总黄酮的静态吸附解吸性能,筛选出最佳树脂AB-8,优化其工艺参数.结果表明,最佳工艺参数为:上样浓度为0.608 mg/mL,上样pH值为6.0,上样体积为5 BV,洗脱液体积为3 BV,洗脱液浓度为60%,上样和洗脱流速同为1.0 m L/min.工艺优化后总黄酮回收率可达71.37%,纯度为79.58%,精制倍数2.95.该工艺能有效分离纯化核桃叶黄酮化合物. To optimize the best macroporous adsorption resin (MAR) through static adsorption and desorption and establish enrichment process of flavonoids from Yili hickory leaves with the concentration of flavonoids as the index. Nine MARs were initially screened for the static adsorption/desorption of flavonoids in the extract, and AB-8 was selected for the best resin. The Results shown that craft parameters of the best resin were optimized and the re- sults were as follows: initial concentration was 0.608 mg/mL, the adsorption of the flavonoids on the resin was effec- tively completed by 5 BV of sample solution of pH 6 with a flow rate of 1 mL/min. The dsorption of the compounds from the resin was best achieved by using 3 BV of 60% ethanol in water at a flow rate of 1 mL/min. After one run of adsorption and desorption, the recovery rate reached up to 71.37%, the purity was 79.58%, the purified multiples of flavonoids was 2.95.
