摘要
目的 :研究瓜蒌皮对高血脂合并急性心肌缺血(HL-AMI)大鼠氧化应激状态的动态调节作用,从调整异常氧化应激状态的角度揭示该药材干预冠心病(CHD)痰浊壅塞证的机制。方法 :雄性Wistar大鼠随机分为对照组(Ctrl)、HL-AMI组、瓜蒌皮干预组(Trich)。首先采用高脂乳剂灌胃,造成高脂血症(HL)大鼠模型,再结扎冠状动脉左前降支制造急性心肌缺血(AMI)模型。瓜蒌皮水煎液(20 g/kg)灌胃给药。上述每组大鼠再分为7个亚组。各亚组分别于冠脉结扎手术后的1-7 d分批处死,腹主动脉取血。生化试剂盒检测血浆中超氧化物歧化酶(SOD)、脂质过氧化物(LPO)、过氧化氢酶(CAT)、总抗氧化能力(T-AOC)和氧化型低密度脂蛋白(ox-LDL)的活性或含量。绘制AMI后7 d内上述指标动态变化曲线。结果 :AMI手术后,HL-AMI组大鼠血浆的SOD、T-AOC及CAT水平显著低于对照组大鼠;而LPO及ox-LDL含量明显升高。灌服瓜蒌皮水煎液的大鼠,其血浆上述氧化应激指标水平的异常变化得到了显著逆转(P〈0.05或P〈0.01)。结论 :对于HL-AMI大鼠,瓜蒌皮可显著改善其AMI后异常的氧化应激状态,较强的抗氧化活性可能是该药材防治CHD痰浊壅塞证的机制之一。
Objective :To study the dynamic regulation function of Pericarpium Trichosanthis on the oxidative stress rats with hyperlipemia and acute myocardial ischemia(HL-AMI),and reveal the mechanism that the medicine intervene the coronary heart disease(CHD)phlegm dampness syndrome from the angle of adjusting the abnormal oxidative stress. Methods :Male Wistar rats were randomly divided into control group(Ctrl),HL-AMI group and Pericarpium Trichosanthis(Trich). First,high-fat emulsion was used to induce hyperlipidemia(HL)rat model,and then the we ligated the rats' left anterior descending coronary artery to make acute myocardial ischemia(AMI)model. All of the rats were given the Trichosanthes Kirilowii Maxim decoction(20 g/kg)treatment. The rats in each group were subdivided into 7 subgroups. Each subgroup was killed in batches 1-7 days after coronary artery ligation,and removing the blood from the abdominal aorta.We tested the activity or content of superoxide dismutase(SOD),lipid peroxide(LPO),catalasein plasma(CAT),total antioxidant capacity(T-AOC)and oxidized low density lipoprotein(ox-LDL)in plasma by biochemical kit. We plotted the dynamic change curves of the above indicators within 7 days after AMI drawing. Result :After AMI,the plasma levels of SOD,T-AOC and CAT in the HL-AMI group were significantly lower than those in the control group,but the content of LPO and ox-LDL increased obviously. After irrigated with Trichosanthes kirilowii Maxim decoction,the abnormal changes in the level of plasma oxidative stress in rats were significantly reversed. Conclusion :In HL-AMI rats,Pericarpium Trichosanthis can significantly improve the abnormal oxidative stress after AMI. Strong antioxidant activity may be one of the mechanisms of the drug prevention and treatment of CHD phlegm syndrome.
