芪黄补肺汤(原防哮汤)抑制哮喘大鼠气道重塑及对细胞色素C影响 被引量:5

Study of Qihuang Bufei Decoction (Fangxiao Decotion)on Airway Remodeling and Cytochrome C in Asthmatic Rats

摘要 目的 :观察芪黄补肺汤抑制SD哮喘大鼠气道重塑的疗效,初步揭示其通过激活细胞色素C(Cyt-C)的释放,促进气道平滑肌细胞(ASMC)凋亡的机制。方法 :采用随机原则将40只SD大鼠分为空白对照组、阳性对照组、顺尔宁组、芪黄补肺汤组。采用鸡卵蛋白(OVA)致敏后,雾化激发12周制备哮喘模型。于第15天起,灌胃治疗12周。HE染色法观察气道病理切片;图像分析软件测量支气管壁总面积(Wat)、支气管平滑肌面积(Wam)等;细胞计数器检测肺泡灌洗液(BALF)中白细胞(WBC)数及嗜酸性粒细胞(EOS)百分比,ELISA法检测ASMC胞内中Cyt-C含量,Tunel法检测ASMC凋亡指数。结果 :与阳性对照组相比,芪黄补肺汤组、顺尔宁组大鼠在气道炎性细胞浸润、气道平滑肌(ASM)层增厚、支气管平滑肌厚度(Wam/Pbm)、支气管壁厚度(Wat/Pbm)、BALF中的WBC数及EOS百分比方面显著降低(P〈0.01),芪黄补肺汤组与顺尔宁组无明显差异(P〉0.05);芪黄补肺汤组、顺尔宁组ASMC凋亡指数和Cyt-C含量较阳性对照组均有上调(P〈0.01),芪黄补肺汤组优于顺尔宁组(P〈0.01)。结论:芪黄补肺汤能够抑制哮喘模型大鼠气道重塑,其机制可能通过激活凋亡蛋白Cyt-C促进ASMC凋亡而实现。 Objective :To observe the therapeutic effect of Qihuang Bufei Decoction on airway remodeling in asthmatic rats with SD,to reveal the mechanism of apoptosis of airway smooth muscle cells(ASMC)promoted by the release of cytochrome C(Cyt-C). Methods :Using the principle of random 40 SD rats were randomly divided into blank negative control group,the positive control group,montelukast sodium intervention group,Qihuang Bufei Decoction intervention group. The model of asthma was induced by ovalbumin(OVA)sensitization and atomization for 12 weeks. In fifteenth days,gavage treatment for 12 weeks. HE staining was used to observe the pathological sections of the airway,and the total area of the bronchial wall(Wat)and the area of bronchial smooth muscle(Wam)were measured by image analysis software;The number of white blood cell(WBC)and eosinophils(EOS)in bronchoalveolar lavage fluid(BALF)were detected by cell counter. The content of Cyt-C in ASMC cells was detected by ELISA method. The apoptosis index of ASMC was detected by Tunel assay. Results :Compared with the positive control group,Qihuang Bufei Decoction group,montelukast group rats in airway inflammation cell infiltration of airway smooth muscle(ASM),the thickness of bronchial smooth muscle layer(Wam/Pbm),bronchial wall thickness(Wat/Pbm),WBC and EOS in percentage of BALF decreased significantly(P〈0.01),Qihuang Bufei Decoction group and montelukast group had no significant difference(P〈0.05);Qihuang Bufei Decoction intervention group and montelukast intervention group ASMC apoptosis index and Cyt-C content compared with the positive control group were increased(P〈0.01),Qihuang Bufei Decoction group was better than the effect of montelukast group. Conclusion :Qihuang Bufei Decoction can inhibit the airway remodeling in asthmatic rats,and its mechanism may promote the apoptosis of ASMC by activating the apoptotic protein Cyt-C.
