摘要
根据实验目的选择合适的模型,是研究疾病发病机制和防治疾病的重要平台和手段。类风湿性关节炎是一种以关节慢性炎症为主要表现的自身免疫性疾病,最常用的动物模型有佐剂性关节炎(AIA)和胶原诱导性关节炎(CIA)。该文从病因学、制备方法、发病特点及发病机制等方面对这两种动物模型进行比较,发现CIA模型病理机制涉及细胞免疫和体液免疫,AIA模型则主要表现为T细胞介导的细胞免疫反应。因此,AIA适用于T细胞分子机制和疗法的研究,而CIA可用于类风湿性关节炎细胞和体液免疫机制研究,该模型的免疫学特征与人类更为相似。该文基于两种模型各自的优缺点,根据研究目的选择适宜的模型,研究类风湿性关节炎的发病机理进而指导临床。
The selection of proper model on the basis of the study objectives is a critical success factor to elucidate the pathogenesis and propose new strategies for preventive,early and curative treatments of certain disease. Rheumatoid arthritis(RA)is a systemic autoimmune disease which is characterized by hyperplasia of synovial tissues. The most common animal models for RA include adjuvant-induced arthritis(AIA)and collagen-induced arthritis(CIA). This review focuses on the comparison of the etiology,modeling methodology,morbidity characteristics and pathogenesis of the two models. It was found that the T cell-mediated cellular immune response was involved in AIA,while CIA processed both cellular and humoral immune response. Hence,AIA model is recommended for the study of T cell-mediated molecular mechanisms and related therapies,while CIA model,which has more similarities with that of human's RA,can be used for the research of both cellular and humoral immune mechanism. In view of the advantages and disadvantages of these two models,the selection of suitable animal model according to diverse research purposes could provide reliable evidence for pathogenesis research and clinical guidance of RA.
