摘要
目的探究并分析甲钴胺联合脑苷肌肽对有机磷中毒迟发性神经病大鼠脑保护及血清神经元特异性烯醇化酶(NSE)mRNA表达的影响。方法选取健康成年Wistar大鼠120只,按照随机数字表法将其分为对照组和观察组,每组60只,对照组大鼠又随机分为未造模组、造模后3 h组(氧化乐果灌胃80 mg/kg,造模后3 h处死)、造模后6 h组(氧化乐果灌胃80 mg/kg,造模后6 h处死),每组20只。观察组大鼠又随机分为未造模组、造模后3 h组(氧化乐果灌胃80 mg/kg+甲钴胺+脑苷肌肽注射,造模后3 h处死)、造模后6 h组(氧化乐果灌胃80 mg/kg+甲钴胺+脑苷肌肽注射,造模后6 h处死),每组20只。观察并记录各组大鼠超氧化物歧化酶(SOD)、丙二醛(MDA)、NSE mRNA水平。结果经重复方差分析,两组大鼠造模前后SOD水平差异显著(P<0.05),两组大鼠造模前SOD水平比较无显著差异(P>0.05),随着造模时间的进行,大鼠SOD水平有所降低,但观察组大鼠降低程度明显低于对照组(P<0.05);经重复方差分析,两组大鼠造模前后MDA水平差异显著(P<0.05),两组大鼠造模前MDA水平比较无显著差异(P>0.05),随着造模时间的进行,两组大鼠MDA水平有所升高,但观察组大鼠升高程度明显低于对照组(P<0.05);经重复方差分析,两组大鼠造模前后NSE mRNA水平差异显著(P<0.05),两组大鼠造模前NSE mRNA水平比较无显著差异(P>0.05),随着造模时间的进行,两组大鼠NSE mRNA水平有所升高,但观察组大鼠升高程度明显低于对照组,差异具有统计学意义(P<0.05)。结论甲钴胺联合脑苷肌肽能有效降低有机磷中毒大鼠脑组织氧化应激水平及血清NSE mRNA表达,具有一定的脑保护作用。
Objective To explore and analyze effect of Mecobalamin combined with GM1 on brain protection and expression of serum NSE mRNA in rats with organophosphorus poisoning delayed neuropathy. Methods 120 healthy adult Wistar rats (clean grade,weight,180 ~ 240 g) were randomly divided into control group and observation group,60 rats in each group. The control group of rats were randomly divided into un-treated group,3 h treatment group (Omethoate intragastric administration of 80 mg/ kg after treatment 3 h killed),after 6 h treatment group (Omethoate intragastric administration of 80 mg/ kg,6 h after treatment,20 rats in each group were sacrificed). The observation group were ran-domly divided into control group,3 h treatment group (Omethoate intragastric 80 mg/ kg + mecobalamin injection after 3 h treatment were the oth-ers,),after 6 h treatment group (80 mg/ kg + Omethoate intragastric administration of mecobalamin injection of 6 h after treatment were encepha-lon,20 rats in each group). The levels of SOD,MDA,NSE and mRNA in each group were observed and recorded. Results Through the analy-sis of variance,the difference of SOD level before and after was significant (P 〈 0. 05),SOD level in two groups before treatment had no signifi-cant difference (P 〉 0. 05),with the treatment time,rats SOD level decreased,but that in the observation group decreased significantly lower than that in the control group (P 〈 0. 05)by the analysis of variance;differences in the level of MDA in two groups before and after treatment were obvious (P 〈 0. 05),while MDA level before treatment in two groups had no significant difference (P 〉 0. 05),with the treatment time, rats MDA levels in the two groups increased,but that in the observation group rats increased significantly lower than that in the control group (P〈 0. 05);and the difference of NSE mRNA levels before and after treatment were significant (P 〈 0. 05),NSE mRNA levels before treatment in the two groups had no significant difference (P 〉 0. 05),with the processing time,the levels of NSE and mRNA in two groups were increased, but the degree of increase in the observation group was significantly lower than that in the control group (P 〈 0. 05),which was statistically signif-icant. Conclusion Mecobalamin combined with GM1 can effectively reduce the the oxidative stress levels and the expression of serum NSE mR-NA in rats with organophosphorus poisoning delayed neuropath,and it has a certain protective effect.
