芒果苷对脑性瘫痪模型大鼠神经运动功能及皮质区神经细胞凋亡的影响 被引量:4

摘要 目的探索芒果苷(MAN)对脑性瘫痪模型大鼠神经运动功能及脑皮质区神经细胞凋亡的影响。方法建立大鼠脑性瘫痪模型后,60只大鼠随机分为5组:假手术组,模型组,芒果苷低浓度组,芒果苷中浓度组和芒果苷高浓度组。芒果苷组在造模后分别给予10 mg/kg、25 mg/kg、50 mg/kg芒果苷治疗,于造模后第1、15和30天进行神经运动功能(BBB)评分,30 d后取出各组大鼠脑组织皮质区进行HE染色和caspase-3、Bax蛋白免疫组织化学染色,以及通过Western blotting检测Bax、Bcl-2蛋白表达。结果 BBB评分显示,第1天模型组和各浓度芒果苷组大鼠BBB评分无显著差异(P>0.05),第15和第30天,芒果苷组BBB评分显著高于模型组,并呈剂量依赖趋势(P<0.05);免疫组织化学染色显示,芒果苷组caspase-3、Bax蛋白平均光密度显著低于模型组,并呈剂量依赖趋势(P<0.05);Western blotting结果显示,芒果苷组Bax蛋白灰度值显著低于模型组(P<0.05),而Bcl-2蛋白灰度值显著高于模型组,并呈剂量依赖趋势(P<0.05)。结论芒果苷能显著改善脑性瘫痪大鼠神经运动功能,抑制脑皮质区神经细胞凋亡,并其作用呈剂量依赖性。 Objective To explore the effect of mangiferin (MAN)on the nerve motor function and cortical neuronal apoptosis in cerebral palsy rats. Methods Cerebral palsy model was made by ligating the left common carotid artery. 60 SD rats were divided into 5 groups:the sham group,Model group,MAN (10 mg/ kg)group,MAN (25 mg/ kg)group and MAN (50 mg/ kg)group. BBB score was valued in the 1st,15th and 30th day atfter surgery. In the 30th day,all brain tissue was collected,and immunohistochemical staining was made to value the caspase - 3 and Bax protein expression,meanwhile,the relative protein expression of Bax and Bcl - 2 were detected by western blotting. Results BBB score showed that there was no significant difference between the MAN group and model group (P 〉 0. 05),and in the 15th and 30th days,BBB score in MAN group was markedly higher than that in model group (P 〈 0. 05). Immunohistochemical staining showed that,the average optical density of caspase - 3 and Bax in MAN were effectively lower than those in model group (P 〈 0. 05). Western blotting showed that the grayscale value of Bax in MAN group was significantly lower than that in model group (P 〈 0. 05),while,the Bcl - 2 was in contrast (P 〈 0. 05). Conclusion Mangiferin can effectively improve the nerve motor function and suppress the cortical neuronal apoptosis in cerebral palsy rats,and its effect was dose - dependent.
