电针曲池、足三里对SHR“两峰”血压、血清褪黑素及5-羟色胺的影响 被引量:9

Observation of Electroacupuncture at Zusanli ( St36) and Quchi ( Li 11) on Two Peaks Blood Pressure and Serum Melatonin and Serotonin in Spontaneously Hypertensive Rats

摘要 目的:观察辰时电刺对自发性高血压大鼠"两峰"血压和血清褪黑素、5-羟色胺水平的影响,探讨其作用时效规律。方法:24只11周龄雄性自发性高血压大鼠(SHR)随机分为两组:针刺组和模型组,每组12只。另取12只WKY大鼠作为正常组。3组大鼠按时辰分为辰、酉两个亚组,每个亚组6只。针刺组于辰时给予电针干预,留针15 min;模型组与正常组在辰时接受捆绑刺激。干预4周后,按时辰分组测量血压,处死后采血。观察治疗后不同组别两个时辰收缩压、舒张压变化,以及褪黑素、5-羟色胺含量变化。结果:干预前模型组、针刺组大鼠尾动脉血压高于正常组(P〈0.05),针刺组与模型组血压和体质量差异无统计学意义(P〉0.05)。干预4周后:(1)辰时:在血压方面,针刺组和模型组SBP、DBP均高于正常组同期(P〈0.05),针刺组低于模型组(P〈0.05);在MT含量方面,针刺组、正常组与模型组有差异(P〈0.05),针刺组与正常组无差异(P〉0.05);在5-羟色胺含量方面,针刺组、模型组与正常组有差异(P〈0.05),针刺组与模型组有差异(P〈0.05)。(2)酉时:在血压方面,针刺组和模型组SBP均高于正常组同期(P〈0.05),针刺组低于模型组(P〈0.05);正常组的DBP低于针刺组和模型组同期(P〈0.05),针刺组和模型组无差异(P〉0.05);在MT含量方面,针刺组与正常组、模型组有差异(P〈0.05),正常组和模型组无差异(P〉0.05);在5-羟色胺含量方面,3组均无差异(P〉0.05)。结论:辰时针刺可以降低SHR"双峰"血压,但以辰时降压效果最佳,其降压效应可能与调节血清MT、5-羟色胺水平有关。 Objective: To observe the effects of electroacupuncture at Zusanli( ST36) and Quchi( LI 11) in 7 a. m to9 a. m on two peaks blood pressure and serum melatonin and serotonin in two peaks on spontaneously hypertensive rats,and its time-effects rules. Methods: Twenty-four male spontaneously hypertensive rats( SHR) aging 11 weeks were randomly divided into acupuncture group and model group,12 rats in each group. The other 12 male WKY rats were selected as control group. Rats in each group were divided into Chen( 7 a. m.-9 a. m.),You( 5 p. m.-7 p. m.) subgroups,6 rats in each group. The acupuncture group received electroacupuncture at 7 a. m-9 a. m for 15 minutes. The model and control groups received binding during 7 a. m-9 a. m. Four weeks after intervention,the blood pressure was measured in all rats according the subgroups,and then rats were executed and blood was collected. The systolic pressure(SBP),diastolic pressure(DBP) and the content of melatonin(MT) and serotonin(5-HT) of different groups after intervention during different time were observed. Results The rats arterial pressure before intervention in acupuncture and model groups were higher than those in control group( P 〈 0. 05),and there were no statistical differences among acupuncture and model groups( P 〉 0. 05). Four weeks after intervention:(1) During 7 a. m-9 a. m,the SBP and DBP in acupuncture and model groups were higher than those in control group( P 〈 0. 05),and the acupuncture group’s was lower than that in model group( P 〈 0. 05). The content of MT in acupuncture and control groups were obvious higher than that in model group( P 〈 0. 05),and there were no statistical differences among acupuncture and control groups(P 〉 0. 05). The content of 5-HT in acupuncture and model groups were higher than that in control group( P 〈 0. 05),and the acupuncture group’s were higher than that in model group( P 〈 0. 05).(2) During 5 p. m-7 p. m,the SBP in acupuncture and model groups were higher than those in control group( P 〈 0. 05) and the acupuncture group was lower than that in model group( P 〈 0. 05). The DBP in control group was lower than those in acupuncture and model groups( P 〈 0. 05) and there were no statistical differences among acupuncture and model groups( P 〉 0. 05). The content of MT in acupuncture group was higher than that in control and model groups( P 〈 0. 05) and there were no statistical differences among control and model groups( P 〉 0. 05). The content of 5-HT had no statistical differences at three groups( P 〉 0. 05). Conclusion: Electroacupuncture at Zusanli( ST36) and Quchi( LI 11) during 7 a. m-9 a. m can reduce the two peaks blood pressure in SHR,with best effects during 7 a. m-9 a. m. Its antihypertensive effects may be related with the regulation of MT and 5-HT contents.
