苗药金茵化石方抗豚鼠胆色素结石性胆囊炎的实验研究 被引量:1

摘要 目的观察苗药金茵化石方对实验性豚鼠胆色素结石性胆囊炎的影响。方法将120只健康豚鼠按照随机数字表法分为空白对照组,模型对照组,消炎利胆片组,苗药金茵化石方低、中、高剂量组,每组20只。除空白对照组外,其余5组复制胆色素结石性胆囊炎模型,造模结束后,空白对照组和模型对照组蒸馏水灌胃;消炎利胆片组采用消炎利胆片0.25 g/kg灌胃,苗药金茵化石方低、中、高剂量组分别采用3.80 g/kg、7.60 g/kg、15.30 g/kg灌胃,各组连续灌胃4周。给药结束后2 d,收集胆汁共2 h测量胆汁量;采集血样标本进行血浆白细胞(WBC)计数,采取胆囊组织测定胆囊组织匀浆上清液IL-2、IL-15和TNF-α含量及检测胆囊组织中TNF-αm RNA表达、组织学检查和胆囊超微结构检查。结果与模型对照组比较,苗药金茵化石方低、中、高剂量组豚鼠胆色素结石性胆囊炎胆汁量明显增加;血浆中白细胞含量及血清和胆囊组织中IL-2、IL-15、TNF-α含量和TNF-αm RNA表达明显降低(P<0.05)。结论苗药金茵化石方具有增加豚鼠胆色素结石性胆囊炎胆汁量,降低血浆中白细胞含量,降低血清和胆囊组织中IL-2、IL-15、TNF-α含量和TNF-αm RNA表达的作用,对豚鼠胆色素结石性胆囊炎具有治疗效果。 Objective To observe the effect of Miao medicine Jinyin stonework on experimental guinea pig bile pigment calculus eholecystitis.nethods 120 healthy guinea pigs were randomly divided into blank control group, model control group, Xiaoyan tablets group, Miao medicine Jin Fang Fang low, medium and high dose group, 20 rats in each group.Except control group, other 5 groups according to the replication of pigment gallstone cholecystitis model.After the modeling, control group and model control group with distilled water; Xiaoyanlidanpian group using 0.25 g/Kg dose administered, Miao medicine Jinyin fossils of low medium and high dose group respectively by 3.80 g/Kg 15.30 g/Kg, 7.60 g/Kg, dose administered,were administered orally for 4 weeks. After medication for 2 days, a total of 2 hour to measure the amount of bile bile collection; blood samples were collected plasma samples from white blood cell (WBC) count and determination of serum IL-2, IL- 15 and TNF- α levels ; take measurement of gallbladder gallbladder tissue homogenate supernatant of IL-2,IL-15 and TNF-α gallbladder tissue and detection of TNF-α mRNA expression histological examination of gallbladder and uhrastructural were detected.Results Compared with the model group,Miao medicine Jinyin fossils of low medium and high dose group in guinea pigs with pigment gallstone cholecystitis bile was increased ; IL-2,IL-15,TNF-α significantly decreased the content of TNF-α mRNA expression in plasma content of leucocyte and serum and gallbladder tissue(P〈0.05 ).Conclusion Miaoyao Jinyin fossils has increased in guinea pig bile pigment calculus cholecystitis bile volume, decreased white blood cell content in plasma,IL-2,IL-15,TNF-α content reduce the effects of and TNF- α mRNA expression in serum and gallbladder tissue, has therapeutic effect on guinea pigs with pigment gallstone.
