人肺癌细胞特异性结合肽的筛选与验证

摘要 目的筛选人肺癌细胞特异性结合肽,为肿瘤诊断试剂及肿瘤药物的研制提供依据。方法以正常人支气管上皮细胞16HBE为吸附细胞,人肺癌细胞A549为靶细胞,利用噬菌体展示肽库经4轮差减吸附淘筛,采用细胞ELISA鉴定噬菌体与肺癌细胞结合的特异性并测序获得多肽序列。结果经4轮筛选噬菌体富集率逐级递增,共挑选394株噬菌体利用ELISA验证,将特异性较好的5株噬菌体克隆基因测序,获得重复率最高的序列(4/5)。结论利用噬菌体展示技术筛选出了与人肺癌细胞癌特异性结合的小分子多肽,为肺癌患者的早期诊断和靶向治疗奠定了基础。 Objective To obtain the peptides specifically binding to human lung cancer cells for developing the diagnosis assay and therapic drug of cancer. Methods Phage display peptide library was panned for 4 rounds with lung epithelial cell 16HBE negative absorbing and lung cancer cell A549 targeting binding. Binding activity was detected with whole-cell ELISA and each clone was identified with gene sequencing. Results Positive clones were gradually enriched after every panning. Five of 394 clones with higher specificity were sequenced, 4 of their sequences were the same. Conclusion Peptides specifically binding to human lung cancer cells are selected, and they provide the base to the early diagnosis and targeting therapy for tumor patients.
