摘要
目的:探索一种适合利用气相色谱法检测粪便中短链脂肪酸(short chain fatty acid,SCFA)的前处理方法。方法:改变粪便样品前处理中所涉及的稀释液种类、稀释液体积、离心转速等实验条件,利用气相色谱法检测不同前处理的粪便样品中SCFA的种类和含量,通过加标回收实验对选择的前处理方法进行验证。结果:选择8 m L酸稀释液处理样品,15 000 r/min离心20 min作为样品的前处理方法,色谱图显示各种SCFA峰形较好,达到基线分离。加标回收率在83.75%~94.95%,符合国家标准。结论:实验中所选择的前处理方法切实可行,适用于粪便中SCFA分析。
Objective:To explore a pretreatment method of the detection of short chain fatty acid (SCFA) in feces by gas chromatography (GC). Methods:The types and contents of SCFA in different pretreated stool samples were detected by GC in the experimental conditions of dilution type, dilution concentration and centrifugal speed in the pretreatment of fecal samples. The pretreatment method was validated by spike recovery experiment. Results:The samples were treated with 8ml acid dilution and centrifuged at 15,000r / min for 20 min. The chromatograms showed that the SCFA peaks were good and the baseline was separated. The recoveries were in the range of 83.75 % to 94.95 %, in line with national standards. Conclusion :The pretreatment method chosen in this experiment is practical and suitable for SCFA analysis in feces.
