牦牛和普通牛CAPN4基因内含子6多态性分析 被引量:1

摘要 本研究采用PCR-SSCP技术和DNA测序法对大通牦牛、天祝白牦牛、甘南牦牛、普通牛4个群体CAPN4基因第6内含子的SNP多态性进行检测,并研究该基因在牦牛和普通牛群体中的遗传特征。试验所得表明:牦牛和普通牛CAPN4基因第6内含子表现出丰富的遗传多样性。CAPN4基因第6内含子检测到5种等位基因A-E,其中等位基因D和E仅存在于普通牛群体中。除天祝白牦牛外,等位基因B频率在其他3个牛群体中均在43%以上为优势等位基因。普通牛PIC<0.25为低度多态外,3个牦牛群体PIC>0.5为高度多态。 In this study, we detected the polymorphism of SNPs of intron 6 in CAPN4 gene in Datong Yak,Tianzhu White Yak, Gannan Yak and cattle by PCR-SSCP and DNA sequencing in order to understand the genetic characters of this gene in yak and cattle. The results showed that the 6 intron of CAPN4 gene in yak and cattle had abundant genetic diversity. 5 A-E alleles were detected in the 6 intron of CAPN4 gene, among which the allele D and E were only found in the common cattle population. Moreover, Allele B was the dominant allele in other three groups of cattle with the allele frequency of more than 43% except for in Tianzhu White Yak. The polymorphism of cattle was low(PIC〈0.25), while the polymorphism of other three yak populations were high(PIC〉0.5).
