摘要
蒙古沙冬青(Ammopiptanthus mongolicus(Maxim)Cheng.f)是中国西北荒漠区唯一的常绿阔叶灌木,具有很强的抗寒和抗旱等耐逆特性。本研究以蒙古沙冬青为研究材料,从中分离到1个编码MYB类转录因子基因,命名为Am MYB-like。该序列长度为1 678 bp,含有一个由1 557 bp组成的开放阅读框(open reading frame,ORF),编码518个氨基酸,具有R2和R3保守结构域,属于R2R3-MYB转录因子。荧光定量PCR分析结果表明,Am MYB-like在叶片中主要参与干旱胁迫应答,在根中主要参与低温及机械胁迫应答。构建了p PZP212-Am MYB-like植物表达载体,为进一步研究Am MYB-like的功能奠定了基础。
Ammopiptanthus mongolicus, has very strong cold resistance and drought tolerance and is only evergreen broad-leaf shrub in the northwest desert of China. In this research, a MYB transcription factor gene was isolated from A. mongolicus, and named as Am MYB-like. The full length of the Am MYB-like c DNA was 1 678 bp,including a single 1 557 bp opening reading frame(ORF) which encoded a 518-amino acid peptide. Am MYB-like contained R2 and R3 conserved domains and belonged to the R2 R3-MYB subfamily. Quantitative real-time PCR(q RT-PCR) analyses revealed that Am MYB-like was up-regulated expressing in leaves under drought stresses, but it mainly induced low temperature and mechanical stress in roots. p PZP212-Am MYB-like expression vector was constructed, and it laid the foundation for further research of Am MYB-like.
