摘要
综述近10多年硒蛋白P(SEPP1)基因与肿瘤的研究进展,主要从血SEPP1蛋白水平与肿瘤发病风险的相关性、血DNA中SEPP1基因多态性与肿瘤遗传易感性、组织中SEPP1基因或蛋白表达变化及机制等三方面进行归纳。首先,血SEPP1蛋白水平低,推测与肿瘤慢性应激条件下肝脏及相关组织该基因表达受到抑制有关,但血SEPP1蛋白主要来源于肝脏分泌,与肿瘤组织本身直接关系并不大;其次,多种肿瘤发生后,SEPP1基因单核苷酸多态性(SNP)多见,可能会在一定程度上使其表达受限,从而与血中蛋白水平变化呈现一致性,这可能属于非特异性改变,但同时也能反映部分基因易感人群特征。再次,肿瘤组织中SEPP1基因或蛋白表达以下降居多,机制多围绕抗氧化展开,但部分机制仍待阐明。此外在后续研究中,可尝试关注SEPP1与免疫、肿瘤细胞的侵袭和转移等诸方面。
This paper reviewed Seppl and cancer in more than one decade. Three aspects were included: the correlation between SEPP1 protein level in blood and the risk of cancer; SEPP1 polymorphism, which from blood DNA sample, and genetic susceptibility to cancer; gene expression changes of SEPP1 in tissues and the underlying mechanism. The result is as follow. Firstly, In the case of low Seppl protein in blood, it can be speculated that gene expression is inhibited in liver and related tissues under chronic stress conditions, while SEPP1 in blood is mainly secreted by liver and not directly related to tumor tissue itself. Secondly, after occurrence of tumor, multiple SNPs appeared in SEPP1 also may limit its expression, which is accord with protein changes in blood; the above may be a nonspecific change and also reflect the characteristics of some gene susceptible population. The third, mechanism of low expression in tumor tissues is mostly around antioxidant while needs further research; Besides, aspects from the relation between SEPP1 and immune, tumor cell invasion, and metastasis can be considered in follow-up research.
