摘要
目的改良经典的烟熏+冷风+LPS(脂多糖)建立"肺气虚外感"大鼠模型的方法 ,减轻对模型大鼠呼吸道的机械损伤及感染率。方法采用"烟熏+冷风刺激+改良经口气管注射LPS"的方式,建立"肺气虚外感"大鼠模型。造模过程中,监测模型大鼠一般情况和体重增长的变化,并与正常大鼠进行对比。于注射LPS后第7 d,处死实验鼠,取肺、脾、胸腺称重,比较模型大鼠与正常大鼠肺、脾、胸腺脏器指数的差异;HE染色,观察比较两组肺组织形态学差异。结果与正常组对比,模型组造模5d后,一般情况、体重逐渐变差;肺、脾、胸腺脏器指数均明显下降(P<0.05~P<0.01);HE染色显示,模型组肺组织呈现炎细胞浸润改变及轻度肺气肿征。结论 "烟熏+冷风刺激+改良经口气管注射LPS"的方式能够成功建立"肺气虚外感"大鼠模型,该方法能减轻对大鼠呼吸道的机械损伤,减少术后伤口感染干扰因素,更符合肺气虚外感的病理生理演变过程。
Objective To modify and use the classical combination of cigarette smoke+cold air+lipopolysaccharide(LPS) to build a rat model of "deficiency of lung-Qi with exogenous cause" and reduce mechanical injury to and the infection rate of the respiratory duct of model rats. Rethods The method "cigarette+cold air stimulation+modified orotracheal injection of LPS" was adopted and the rat model "deficiency of ]ung-Qi with exogenous cause" was created. During the-modeling, the changes in general conditions and weight growth of the model rats were monitored and compared with those of the normal rats. Seven days after the LPS injection, the model rats were killed. The lung, spleen and thymus gland were taken out and weighed. The differences among the indexes of lung, spleen and thymus gland were compared between the model rats and the normal rants; the hematoxylin-eosin (HE) staining was done and then the morphological difference of lungs between the two groups were observed. Results Five days after the modeling, the general conditions got worse and the weights decreased in the model rat group; the indexes of lung, spleen and thymus gland dropped obviously (P 〈 0.05-P 〈 0.01 in each case); the HE staining showed inflammatory cell infiltration and slight sign of emphysema in the lung tissue of the model rat group. Conclusion In the method "cigarette smoke+cold air stimulation+modified orotracheal LPS injection", the rat model "deficiency of lung-Qi with exogenous cause" can be successfully established; this method can relieve mechanical injury to the respiratory duct of rat and reduce interference factors that cause post-operative wound infection, and it coincides with the process of pathophysiological evolution of deficiency of lung-Qi with exogenous cause.
