硬皮病患者皮损中Smad3与Smad7 mRNA的表达及检测意义 被引量:1

摘要 目的本次研究的目的是检测硬皮病患者皮损中Smad3和Smad7 mRNA的表达,探讨其在硬皮病发病机制中的作用及临床检测意义。方法本研究共纳入2009年2月—2012年6月我院收治的46例未经治疗的硬皮病(SD)患者作为研究对象。另选取15例同期在我院整形外科进行手术患者的健康正常皮肤作为对照。采用组织原位RT-PCR法检测SD患者皮损及对照组患者正常皮肤中Smad3和Smad7的mRNA表达水平。采用Spearman秩相关性分析对2组患者Smad3与Smad7 mRNA的相关性进行分析。比较不同分型与不同分期SD患者皮损中Smad3与Smad7 mRNA的表达水平。结果与对照组正常皮肤相比,SD患者皮损中Smad3 mRNA的表达量显著升高(P<0.05),而Smad7 mRNA的表达量则显著降低(P<0.05)。相关性分析显示,对照组正常皮肤中Smad3与Smad7的mRNA水平呈正相关(P<0.05),而SD患者的皮损中Smad3与Smad7的mRNA水平差异无统计学意义(P>0.05)。不同分型与分期SD患者皮损中Smad3与Smad7 mRNA水平相比差异无统计学意义(P>0.05)。结论本次研究表明SD患者皮损中Smad3 mRNA的上调与Smad7 mRNA的下调可能参与了其病理发生机制,提示Smad3与Smad7mRNA的检测对SD患者具有一定的诊断与治疗价值。 Objective In order to measure the expression level of Smad3 and Smad7 mRNA in skin lesions of patients with scleroderma and to explore the role of Smad3 and Smad7 mRNA in the pathogenesis of scleroderma. Methods A total of 46 cases of untreated patients with scleroderma(SD) in our hospital from February 2009 to June 2012 were recruited. Another 15 patients with normal skin treated in orthopedics were selected as controls. The expression level of Smad3 or Smad7 mRNA in skin lesions from SD patients and in normal skin from controls were detected by in situ RT-PCR. Spearman correlation analysis was used to evaluate the correlation between Smad3 and Smad7. The expression levels of Smad3 and Smad7 mRNA in SD patients with differently type and stage were compared. Results Compared with controls, the Smad3 mRNA level in scleroderma lesions were significantly increased(P〈0.05), while Smad7 mRNA level were significantly decreased(P〈0.05).Correlation analysis showed Smad3 mRNA level in control group was positive correlated with Smad7 mRNA level(P〈0.05),however there was no correlation between Smad3 and Smad7 mRNA levels in skin lesions of SD patients(P 0.05). No significant differences of Smad3 and Smad7 mRNA levels were found between different types or stages of SD patients(P 0.05). Conclusion Our results showed that Smad3 mRNA up-regulation and Smad7 mRNA down-regulation may be involved in the pathogenesis of scleroderma, indicating that detection of Smad3 and Smad7 mRNA may poses diagnostic and therapeutic value for SD patients.
