白花蛇舌草对肝癌的作用机制研究 被引量:21

摘要 目的:探讨中药白花蛇舌草治疗肝癌的作用机制。方法:建立荷瘤小鼠肝癌模型,将造模成功的15只小鼠随机分为空白对照组、白花蛇舌草组和5-氟尿嘧啶溶液组,每组各5只。空白对照组给予清水灌胃,白花蛇舌草组给予白花蛇舌草注射液,每只50 mg·kg-1,肌肉注射,每周1次,共治疗5次,5-氟尿嘧啶溶液组腹腔注射5-氟尿嘧啶溶液。观察各组荷瘤小鼠肿瘤生长情况,采用ELISA法检测血清中干扰素-γ(interferon-γ,IFN-γ)、白细胞介素-12(interleukin-12,IL-12)、免疫球蛋白G(Ig G)、免疫球蛋白M(Ig M)、甲胎蛋白(alpha fetoprotein,AFP)的分泌水平,采用细胞毒实验MTT检测中药白花蛇舌草对肝癌细胞的抑瘤作用。结果:10%、15%、20%中药白花蛇舌草含药血清组抑制率分别为(14.2±1.42)%,(24.60±3.76)%,(35.35±4.81)%,其中以20%的浓度抑瘤率最高,与其他两组比较,差异具有统计学意义(P<0.05)。空白对照组、白花蛇舌草组、5-氟尿嘧啶溶液组小鼠肝癌的发生率分别为100%、60%、30%,三组间比较,差异有统计学意义(P<0.05)。空白对照组、白花蛇舌草组、5-氟尿嘧啶溶液组小鼠肿瘤的体积分别为(3 042±56)mm3、(1 873±47)mm3、(873±12)mm3,与空白对照组和白花蛇舌草组比较,5-氟尿嘧啶溶液组小鼠肿瘤的体积最小,差异有统计学意义(P<0.05)。与空白对照组,白花蛇舌草组相比,5-氟尿嘧啶溶液组荷瘤小鼠血清中Ig G,Ig M以及INF-γ、IL-12的含量明显升高,差异具有统计学意义(P<0.05),而5-氟尿嘧啶溶液组荷瘤小鼠血清中AFP含量明显降低,差异具有统计学意义(P<0.05)。结论:中药白花蛇舌草通过提高荷瘤小鼠体内Ig G,Ig M以及INF-γ,IL-12的分泌水平,降低AFP含量,增强了机体细胞免疫及体液免疫能力,使机体可有效的识别并清除肿瘤细胞,抑制了肿瘤血管的生成。 Objective : To explore the mechanism of Bat Hua She She Cao ( Herba Hedyotis Diffusae) in treating liver cancer. Meth- ods:Hepatoma model of tumor bearing mice was first established. Then such 15 model mice were randomly divided into the blank control group,Bat Hua She She Cao suspension group,5- fluorouracil solution group, with 5 mice in each group. The ones in tile blank control group were given water gavage,and he ones in the Bat Hua She She Cao group were given intramuscular injection, each 50 mg · kg^-1, once a week. The ones in the 5-fluorouracil solution group were given intraperitoneal injection. The tumor growth was observed in mice bearing tumor. ELISA method was used to detect serum interferon-y, (IFN-y) ,interleukin-12 ( IL- 12) ,immunoglobulin G (IgG) ,immunoglobulin M ( lgM ) , alpha fetoproteiu (AFP) secretion level. The inhibitory effect of Bat Hua She She Cao on liver-cancer cells was detected by using cytotoxicity test of MTT. Results:The inhibition rates of 10% , 15% , 20% of Bat Hua She She Cao medicated serum groups were ( 14.2 ± 1.42) % and ( 24.60 ± 3.76) % and ( 35.35 ± 4.81 ) % respectively ,of which 20% of the concentration inhibition rate was highest, compared with other two groups, the difference was statistically significant (P 〈 0.05 ). The incidences of liver cancer in mice in the blank control group, Bat Hua She She Cao sus- pension gruup,5-fluorouracil solution group were 100% ,60%, and 30% respectively. The difference between the three groups was statistically significant (P 〈 0.05). The volumes of tumor in mice in the blank control group, Bat Hua She She Cao suspen- sion group ,5- fluorouraeil solution group were (3042 ± 56) mm3, (1873 ± 47 )mm3 and (873 ± 12 )mm3 respectively. Compared with the blank control group,and Bat Hua She She Cao suspension group,the volume of the tumor of mice in 5-fluorouraeil solu- tion group was the smallest,and the difference was statistically significant (P 〈 0.05). Compared with the blank control group, and Bat Hua She She Cao suspension group,the nfice serum IgG, IgM, INF-gamma,IL-12 content in 5-fluorouraeil solution group increased obviously, the difference was statistically significant ( P 〈 0.05 ) ; and 5-fluorouraeil solution group in tumor bearing mice serum AFP content decreased significantly, and the diflereuee was statistically significant ( P 〈 0.05 ). Conclusion : By increasing IgG, IgM and the secretion levels of INF-α and IL-12 in tumor bearing mice decreasing the content of AFP, Bat Hua She She Caoe can enhance cellular immunity and humoral immunity, so that the body can effectively identify and remove tumor cells, and inhibit tumor anglogenesis.
