摘要 该研究利用斑马鱼全胚胎原位杂交技术和转基因(VEGFR2:GFP)斑马鱼模式生物,评估miR-15b对斑马鱼早期胚胎发育和后期血管生成的影响。实验结果表明:miR-15b可能是母源性分子,在斑马鱼早期胚胎中高表达,而过表达miR-15b导致斑马鱼血管发育畸形。蛋白免疫印迹和双荧光素酶报告基因检测实验结果显示,miR-15b通过结合VEGFR2 m RNA 3’UTR区降低VEGFR2基因的表达水平,从而调控斑马鱼的血管生成。综上所述,miR-15b在斑马鱼胚胎发育过程中具有重要的作用,特别是对血管系统发育产生影响。该结果可为进一步研究miR-15b在斑马鱼胚胎发育过程的功能提供参考。 In this study, the effects of miR-15b on embryonic development and angiogenesis of zebrafish were studied by using zebrafish whole-embryo in situ hybridization and transgenic (VEGFR2: GFP) zebrafish model. The results showed that miR-15b may be highly expressed in early embryos of zebrafish, and overexpression of miR-15b leads to angiogenic malformations in zebrafish. Western blot and dual-fluorescence reporter system showed that miR-15b regulated the expression of VEGFR2 gene by binding to 3'UTR of mRNA, thus to regulate the angiogenesis of zebrafish. In summary, miR-15b plays an important role in the development of zebrafish embryos, especially in the vascular system. The results provide a theoretical basis for further studies on miR-15b in the embryonic development of zebrafish.
