舍曲林对抑郁模型小鼠海马MicroRNAs水平的影响 被引量:4

摘要 目的探讨舍曲林对抑郁模型小鼠海马MicroRNAs水平的影响。方法选取8~12周的雄性昆明小鼠,分为对照组(A组)、模型组(B组)和造模+治疗组(C组)各8只。A组为5周正常饲养,B组经过5周慢性轻度不可预见性的应激抑郁(CUMS),C组经过3周CUMS随后进行舍曲林腹腔注射2周。使用体质量测量和糖水偏好实验评估小鼠抑郁程度,并检测小鼠海马MicroRNAs水平。结果入组后第3、4、5周末,B组和C组体质量、糖水偏好指数低于A组(P<0.05)。入组后第4、5周末,C组体质量、糖水偏好指数高于B组(P<0.05)。B组海马miRNA-132和miRNA-18a水平低于A组(P<0.05),miRNA-134和miRNA-124a水平高于A组(P<0.05)。C组海马miRNA-132和miRNA-18a水平高于B组(P<0.05),miRNA-134和miRNA-124a水平低于B组(P<0.05)。结论舍曲林可能通过调节海马MicroRNAs水平发挥抗抑郁作用。 Objective To explore the effects of sertraline to influence level of microRNA expression in mice with depression.Methods A total of 24 mice were randomly divided into group A(8 cases) with normal feed for 5 weeks,group B(8 cases)with chronic unpredictable mild stress(CUMS) for 5 weeks and group C(8 cases) with CUMS for 3 weeks and treatment of sertraline for 2 weeks.They were examined with weight measurement,sucrose preference test and the levels of microRNAs on hippocampus.Results At the 3^(rd),4^(th) and 5^(th) weekend,the weight measurement and sucrose preference index in group B and C were all significantly lower than those in group A(P 0.05).At the 4^(th) and 5^(th) weekend,the weight measurement and sucrose preference index in group C were all significantly higher than those in group B(P 0.05).The levels of miRNA-132 and miRNA-18 a on hippocampus in group B were significantly lower than those in group A(P 0.05),and the levels of miRNA-134 and miRNA-124 a in group B were significantly higher than those in group A(P 0.05).The levels of miRNA-132 and miRNA-18 a on hippocampus in group C were significantly higher than those in group B(P 0.05),and the levels of miRNA-134 and miRNA-124 a in group C were significantly lower than those in group B(P 0.05).Conclusion There may be important antidepressant role for sertraline to adjust the level of microRNAs on hippocampus.
