积雪草酸对脂多糖所致H9C2心肌细胞损伤的改善作用 被引量:7

摘要 目的探讨积雪草酸对脂多糖所致H9C2心肌细胞损伤的改善作用及其作用机制。方法将H9C2心肌细胞分为空白对照组、积雪草酸组、脂多糖组以及脂多糖+积雪草酸组,分别给予磷酸缓冲盐溶液(20μmol/ml)、积雪草酸(20μmol/ml)、脂多糖(1μg/ml)、脂多糖(1μg/ml)+积雪草酸(20μmol/ml)进行干预。干预24 h后检测4组细胞的存活率、肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、IL-6 mRNA表达水平、细胞核内核因子-κB(NF-κB)蛋白表达水平以及超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性、细胞内丙二醛水平。结果积雪草酸可使H9C2细胞存活率上升,并使TNF-α、IL-1β和IL-6的mRNA水平降低,NF-κB核转位明显减少,SOD、GSH-Px活性增加、丙二醛水平下降。结论积雪草酸可通过抑制炎症和氧化应激改善脂多糖引起的心肌细胞损伤。 Objective To investigate the effect of asiatic acid on the improvement of lipopolysaccharide( LPS)-induced injury to myocardial H9 C2 cells. Methods The myocardial H9 C2 cells were divided into blank control group,asiatic acid group and LPS + asiatic acid group,and were given intervention with phosphate buffered saline( PBS)( 20 μmol/ml),asiatic acid( 20 μmol/ml),LPS( 1 μg/ml),and LPS( 1 μg/ml) + asiatic acid( 20 μmol/ml) respectively. After 24 hours of intervention,the survival rate,mRNA expression levels of tumor necrosis factor-α( TNF-α),interleukin( IL)-1β and IL-6,expression levels of endonuclear nuclear factor-κB( NF-κB) protein,activities of superoxide dismutase( SOD) and glutathione peroxidase( GSH-Px),and intracellular level of malondialdehyde( MDA) were detected among the four groups. Results Asiatic acid could increase the survival rate of H9 C2 cells,decrease the mRNA levels of TNF-α,IL-1β and IL-6,reduce the nuclear translocation of NF-κB significantly,increase the activities of SOD and GSH-Px,and decrease the MDA level. Conclusion Asiatic acid can inhibit the LPS-induced myocardial cell injury by suppressing the inflammation and oxidative stress.
