摘要
目的研究血浆游离DNA含量与完整性指数对胰腺癌的诊断价值。方法收集正常人群及胰腺癌患者血液标本,提取基因组DNA,建立游离DNA定量标准曲线;提取血浆游离DNA,用SYBR Green荧光定量PCR法对其进行含量及完整性指数检测,分析2组之间的差异,用ROC曲线判断分析其对胰腺癌的诊断能力。结果胰腺癌组游离DNA浓度及完整性指数均高于对照组,2组血浆游离DNA含量及完整性差异有统计学差异(Z=-2.92,Z=-6.07,P<0.05)。游离DNA浓度及完整性指数对胰腺癌诊断ROC曲线下面积分别为0.669(95%CI:0.563~0.776)、0.806(95%CI:0.723~0.888)。结论血浆游离DNA浓度及完整性指数均对胰腺癌具有一定的诊断价值,完整性指数的诊断能力虽优于游离DNA浓度,但诊断效果并不理想,尚需深入研究更为有效的胰腺癌的诊断方法。
Objective To study the value of plasma free DNA content and integrity index in pancreatic cancer diagnosis.Methods Blood samples were collected from normal and pancreatic cancer patients. Genomic DNA was extracted and the quantitative standard curve was established. Plasma free DNA was extracted,the content of DNA was detected by SYBR Green fluorescence quantitative PCR. And the free DNA content and integrity index between the both groups were analyzed. ROC curves were applied to determine the diagnostic ability of free DNA content and integrity index for pancreatic cancer. Results The concentration and integrity of free DNA in pancreatic cancer group were significantly higher than those in normal group,with the differences statistically significant( Z =-2. 92,Z =-6. 07,P〈0. 05). The area under the ROC curve was 0. 669( 95% CI: 0. 563-0. 776) and 0. 806( 95% CI: 0. 723-0. 888),respectively. Conclusion The plasma free DNA concentration and integrity index have diagnostic value for pancreatic cancer. The diagnostic ability of integrity index is better than DNA concentration,but the diagnostic effect is not ideal. More effective diagnosis method of pancreatic cancer still needs in-depth study.
