摘要 目的:探讨肝细胞癌(HCC)的发生与WIF-1基因启动子甲基化的相关性。方法:通过甲基化特异性PCR(MSP)检测WIF-1基因启动子在76例(实验组)HCC组织中的甲基化率,并随机留取伴肝硬化患者癌旁肝硬化组织标本47例(对照组),分析WIF-1基因启动子甲基化与HCC的相关性。结果:与肝硬化癌旁组织比较,HCC组织WIF-1基因启动子甲基化阳性率明显增高(P<0.05),并且HCC组织中WIF-1甲基化阳性率与甲胎蛋白(AFP)之间存在负相关关系(r=-0.759,P<0.001)。结论:WIF-1基因启动子甲基化与HCC的发生有关;在HCC中WIF-1基因启动子甲基化与AFP浓度呈明显负相关关系。 Objective: To investigate the relationship between WIF-1 promoter methylation and the occurrence of hepatocellular carcinoma(HCC). Methods: Methylation-specific PCR(MSP)was applied to examine the WIF-1 promoter methylation in 76 HCC tissues and 47 paracancerous tissues. The correlation between the WIF-1 gene promoter methylation and HCC was analyzed. Results: The methylation rate of WIF-1 promoter was significantly higher in HCC tissues than that in paracancerous tissues(P〈0. 05). Moreover,WIF-1 methylation rate was negatively correlated with alpha fetoprotein(AFP)in HCC tissues(r=-0. 759,P〈0. 001). Conclusion: WIF-1 promoter methylation was associated with the occurrence of HCC,and was negatively correlated with AFP concentration in HCC.
