摘要
观察白术水提物(Water extract of atractylodis macrocephala,WEA)对血管性痴呆(vascular dementia,VD)大鼠学习记忆障碍的改善作用,初步探讨其药理作用机制。32只健康成年雄性SD大鼠随机分为四组:假手术组、模型组、WEA低剂量组、WEA高剂量组,每组8只。除假手术组外,其余各组大鼠均采用双侧颈总动脉永久阻断法(2-VO)建立血管性痴呆动物模型,14天后,Morris水迷宫实验检测大鼠学习记忆能力,同时测定皮层SOD和MDA含量。结果表明:与假手术组相比,模型组大鼠逃避潜伏期明显延长(P<0.01),在原平台象限停留时间明显缩短(P<0.01),皮层SOD含量明显减少(P<0.01),MDA含量明显增多(P<0.01)。与模型组比较,WEA高剂量组和低剂量组大鼠逃避潜伏期明显缩短(P<0.01),在原平台象限停留时间明显延长(P<0.01,P<0.05),皮层SOD含量显著增多(P<0.05),MDA含量无明显变化。结果提示WEA对VD大鼠学习记忆障碍具有显著改善作用,机制可能与其提高脑组织抗氧化能力有关。
To investigate the effect of water extract of Atractylodis macrocephala on learning and memory impairment in rats with vascular dementia, and to explore its pharmacological mechanism. 32 healthy adult male SD rats were randomly divided into four groups: sham operation group, model group, WEA low-dose group and WEA high-dose group, with 8 rats in each group. All rats in the other groups except the sham operation group were established vascular dementia model with bilateral carotid artery permanent occlusion (2-VO). 14 days later, Morris water maze test was used to detect the learning and memory ability of rats, and the contents of SOD and MDA in cortex were measured simultaneously. The results showed that compared with the sham operation group, the escape latency was significantly prolonged in the model group(P〈0.01), the residence time in the original platform quadrant was significantly shortened(P〈0.01), cortical SOD content was significantly reduced(P〈0.01), MDA content increased significantly(P〈0.01). Compared with the model group, the escape latency of WEA high dose group and low dose group was significantly shortened (P〈0.01), and the residence time in the original platform quadrant was significantly prolonged (P〈0.01,P〈0.05), and the content of SOD in cortex increased significantly(P〈0.05), but the content of MDA had no significant change. The results suggest that WEA can significantly improve learning and memory impairment in VD rats, and the mechanism may be related to enhancing the antioxidant capacity of brain tissue.
