摘要
通过正交试验方法分析提取温度、提取时间、提取液体积及振荡频率4种因素对大豆水溶性蛋白质测定结果的影响,确定大豆水溶性蛋白质最佳提取条件。结果表明,在一定范围内,大豆水溶性蛋白质含量随着提取温度升高、提取时间延长、提取液体积及振荡频率增大而增大,其中振荡频率对水溶性蛋白质含量的影响达到了显著水平。在已处理样品中加入200 mL提取液,于35℃恒温水浴下以180 r/min回旋振荡120 min时,水溶性蛋白质含量最高。该提取工艺高效、快速、经济,为水溶性蛋白的准确测定以及大豆储存品质的准确判断提供参考。
The influence of four factors such as temperature, time, liquid volume and oscillate frequency to thewater–soluble protein content of soybean was analyzed with the orthogonal test method. The optimum extraction conditionsof soybean water soluble proteins were determined. The results showed that the water–soluble protein content increasedwith the rising of temperature, extending time, extraction liquid and oscillate frequency within limits, the effect of oscillatefrequency was significant. As 200 mL extraction liquid was added to the treated samples, the water–soluble protein contentwas the largest under the constant temperature water bath at 35 ℃ with 180 r/min cyclotron oscillation 120 min, whichwere the optimum extraction conditions. The process is efficient, rapid and economical, which provides a reference for theaccurate determination of water soluble proteins and the accurate judgement of soybean storage quality.
