摘要
为制备外源病毒检验用抗鸡新城疫病毒(Newcastle disease virus,NDV)的特异性血清,对300只3~4周龄的SPF鸡进行了基础免疫和加强免疫。最后一次免疫后21 d采血并分离血清,血清经混合、分装、冷冻真空干燥后,对其进行无菌检验、外源病毒检验、剩余水分测定、中和效价测定和特异性检验。结果表明,制备的抗NDV特异性血清无菌检验、外源病毒检验和剩余水分测定均符合现行《中华人民共和国兽药典》规定;血清中和效价为1∶2 089,与NDVLa Sota株毒种中和指数为108.2,与其它禽类病毒的中和指数均不大于10,通过AGP、ELISA、HI等试验确定血清中不含其他禽源外源病毒抗体。该研究为鸡新城疫活疫苗或毒种的外源病毒检验提供了具有良好特异性和高效价的中和用血清。
To prepare the specific antiserum against Newcastle disease virus (NDV)for exogenous virus test, 300 SPF chickens with 3 to 4 weeks of age were inoculated with ND live vaccine and inactivated vaccine. At the 21th day after the last immunity, serums were collected and mixed thoroughly. After lyophilization,the serum was used for sterility test, exogenous virus test, residual moisture test, neutralization titer determination and specific inspection. The results was complied with China Veterinary Pharmacopoeia. The neutralization titer was 1:2 089. The cross neutralization index of the serum induced by NDV La Sota was 108.2, but was below 10 induced by other avian viruses. There were no antibodies against other avian disease viruses in this serum,using APG, ELISA and HI test. It's suggested that antiserum against NDV was prepared with high specificity and neutralization titer for the identify inspection and exogenous virus test of ND live vaccines and virus seeds.
